AB  - Purpose: To compare the clinical performance of a glass hybrid restorative system, EQUIA Forte, with that of a nanohybrid resin composite, Tetric EvoCeram, in two-surface class II cavities. Materials and Methods: This multicenter, randomized controlled clinical study was conducted at four different dental schools. In total, 360 restorations were placed in patients in need of two class-II, two-surface restorations in the molar region of the same jaw. Each patient received one glass hybrid restoration (EQUIA Forte, GC) and one resin composite restoration (Tetric EvoCeram, Ivoclar Vivadent). Two independent evaluators performed a clinical evaluation of each site after 1 week (baseline), 1 year, and 2 years using the criteria of the FDI World Dental Federation (FDI-2). Results: The estimated survival rates at the 2-year recall were 93.6% and 94.5% for EQUIA Forte and Tetric EvoCeram, respectively. There were no significant differences in the survival rates or in any of the evaluated esthetic, functional or biological properties between EQUIA Forte and Tetric EvoCeram restorations (p > 0.05). Conclusion: Both the glass-hybrid restorative system and nanohybrid resin composite showed good clinical performance in moderate to large two-surface class II restorations in a 2-year follow-up.

Mineral trioxide aggregate for the treatment of external root resorption in an avulsed immature tooth: Ten years of follow-up

AB  - Introduction Root resorption may occur as a consequence of avulsion injury and may lead to the progressive loss of tooth structure. The aim was to report the outcome of root resorption treated with mineral trioxide aggregate in a replanted immature permanent incisor after 10 years of follow-up. Case outline This case presents external root resorption that was detected 18 months after the avulsion injury in a nine-year-old child. Apical portion of the canal was filled with mineral trioxide aggregate and the rest of the canal was filled with a canal sealer and gutta-percha. Control examinations were performed six months after the completion of the endodontic treatment and afterwards yearly. The tooth was asymptomatic clinically and radiographs did not show progression of root resorption up to four years of follow-up. Infraposition of the injured tooth was detected five years after the replantation, but without significant radiographic changes, until the eight-year follow-up, when root resorption was detected again. However, the tooth was still hard and symptomless at the 10-year follow-up. Conclusion Mineral trioxide aggregate may have an important role in the preservation of replanted immature teeth for a prolonged period.
AB  - Uvod Resorpcija korena zuba se može javiti kao posledica povrede i može voditi progresivnom gubitku zubnih struktura. Cilj rada je bio da se prikaže klinički ishod replantacije izbijenog stalnog zuba sa nezavršenim rastom korena posle deset godina. Prikaz bolesnika Predstavljen je slučaj eksterne resorpcije korena zuba detektovane 18 meseci posle avulzione povrede kod devetogodišnjeg deteta. Apikalni deo kanala korena zuba je napunjen mineralnim trioksidnim agregatom, dok je ostatak kanala opturiran pastom i gutaperka poenima. Kontrolni pregledi su obavljani šest meseci posle završetka endodontskog lečenja i, posle toga, jednom godišnje. Do četvrte godine praćenja zub je bio bez kliničkih i radiografskih znakova progresije resorpcije korena. Infrapozicija povređenog zuba uočena je pet godina posle replantacije, ali bez značajnih radiografskih promena sve do osme godine praćenja, kada je uočeno napredovanje resorpcije korena. Ipak, posle deset godina zub je i dalje bio klinički bez simptoma. Zaključak Mineralni trioksidni agregat može imati značajnu ulogu u dugotrajnom očuvanju replantiranih zuba sa nezavršenim rastom korena.

Breastfeeding: The perspective of paediatric dentist

AB  - Current recommendations by the European Academy of Paediatric Dentistry, American Academy of Paediatric Dentistry, and International Association of Paediatric Dentistry advocate weaning from breast milk and avoiding unrestricted breastfeeding after the eruption of primary teeth in order to lower the risk of early childhood caries (ECC). However, World Health Organization, American Academy of Paediatrics and nutritional recommendations support exclusive breastfeeding up to six months of age, following continued breastfeeding along with appropriate complementary foods, favouring unrestricted and prolonged breastfeeding even beyond the age of two. The purpose of this review is to discuss current data in the literature regarding the association between breastfeeding and ECC in order to address this problem and to provide consistent recommendations. PubMed search revealed possible link between breastfeeding and ECC, however without evidence strong enough to establish the appropriate oral health preventive recommendation. Having in mind known benefits of breastfeeding, it is advisable to adhere to current paediatric guidelines which promote unrestricted breastfeeding as long as it is mutually desired by mother and child. This recommendation doesn't exclude but complements the prevention and timely treatment of ECC. Furthermore, there is a need to highlight the importance of education of parents and health care providers about the ECC risk factors, identification of initial lesions and consequences. Further research regarding this issue is needed.
AB  - Savremene preporuke Evropske i Američke akademije dečjih stomatologa i Međunarodnog udruženja dečjih stomatologa savetuju postepeni prekid dojenja nakon nicanja mlečnih zuba kako bi se smanjio rizik od nastanka karijesa ranog detinjstva (KRD). Sa druge strane, preporuke Svetske zdravstvene organizacije, preporuke Američke akademije pedijatara i stručnjaka iz oblasti ishrane prepoznaju brojne kratkoročne i dugoročne pozitivne efekte dojenja i podržavaju isključivo dojenje do uzrasta od šest meseci, a zatim postepeno uvođenje čvrste hrane sa nastavkom dojenja uz neograničeno i dojenje na zahtev do druge godine deteta i duže. Cilj ovog preglednog rada je bio da se analiziraju savremeni podaci u literaturi o uticaju dojenja na nastanak KRD, kako bi se doprinelo formiranju jedinstvenog stava i pružila jasna informacija majkama kako prevenirati KRD. Na osnovu pretraživanja Pub Med baze podataka, uočava se da postoji povezanost između dojenja i KRD, ali nije dovoljno argumentovano koje su najbolje mere u prevenciji karijesa. Imajući u vidu poznate pozitivne efekte dojenja, smatra se da je preporučljivo pratiti savremene pedijatrijske preporuke koje savetuju neograničeno dojenje koliko God to uzajamno prija majci i detetu. Ipak, potrebno je imati u vidu neophodnost ranih preventivnih poseta dečjem stomatologu i edukacije zdravstvenih radnika radi adekvatnih i blagovremenih saveta o higijeni usne u duplje i ishrani kako bi se izbegao nastanak karijesa ranog detinjstva i omogućilo blagovremeno dijagnostikovnje inicijalnih lezija. Neophodna su dalja istraživanja u ovoj oblasti.

Imajući u vidu poznate pozitivne efekte dojenja, smatra se da je preporučljivo pratiti savremene pedijatrijske preporuke koje savetuju neograničeno dojenje koliko God to uzajamno prija majci i detetu. Ipak, potrebno je imati u vidu neophodnost ranih preventivnih poseta dečjem stomatologu i edukacije zdravstvenih radnika radi adekvatnih i blagovremenih saveta o higijeni usne u duplje i ishrani kako bi se izbegao nastanak karijesa ranog detinjstva i omogućilo blagovremeno dijagnostikovnje inicijalnih lezija. Neophodna su dalja istraživanja u ovoj oblasti.",

How Much Country Economy Influences ECC Profile in Serbian Children-A Macro-Level Factor Analysis

AB  - Introduction: Serbia has universal health coverage (UHC) for pediatric dental care and similar country distribution for dentists and physicians per 1,000 inhabitants. However, a high prevalence of early childhood caries (ECC) with wide variation across the country was observed in previous studies. This paper aimed to analyze the association between economic and healthcare country macro-level factors with ECC prevalence and treatment. Method: The outcome variables were ECC prevalence and frequency of untreated ECC in 36- to 71-month-olds. Cross-sectional pathfinder survey on a nationally representative sample of children was conducted in order to obtain data. Independent variables included the following: gross domestic product (GDP), social and health care budget beneficiaries' expenditures, local self-government budget, unemployment rate, population density and density of physicians and dentists. Guided by the WHO's Basic Methods for Oral Health Surveys stratified cluster sample, 17 sites were randomly chosen to obtain adequate distribution of data regarding urban, peri-urban and rural areas in each analyzed statistical territorial unit. The variables were analyzed using the independent t-test or Mann-Whitney U test. A probability value of  lt 0.05 was considered significant. Results: The final sample included 864 children aged 36 to 71 months. Observed prevalence of ECC was 41.1%. Although no statistically significant difference was found, children with ECC compared to healthy children were living in parts of the country with averages of approximate to 122(sic) lower GDP per capita, approximate to 4(sic) lower social and health care expenditures per capita, 9 inhabitants per km(2) lower population density, almost 7(sic) per capita lower local self-government budget and a 0.6% higher unemployment rate. Furthermore, although without a statistically significant difference, untreated ECC was associated with approximate to 302(sic) lower GDP per capita, approximate to 12(sic) lower social and health care expenditures per capita, 34 inhabitants per km2 lower population density, almost 20(sic) per capita lower local self-government budget and a 1.7% higher unemployment rate. Conclusions: This study, performed in a nationally representative sample of preschool children, revealed the association of economic macro-level factors with ECC prevalence and its (non-) treatment. Further research on a larger sample is necessary to confirm the results. These findings suggest that most of the public-health efforts regarding prevention and early treatment of ECC should be directed at regions with lower economic performance.

Endodontic treatment of traumatized teeth with chronic periapical lesions using antibiotic paste and mineral trioxide aggregate obturation: A preliminary study

AB  - Introduction/Objective the purpose of this study was to assess effectiveness of endodontic root canal procedures in traumatized permanent teeth with necrotic pulps and chronic periapical lesions after definitive obturation with mineral trioxide aggregate (MTA) products. Adobe Photoshop CS (San Jose, CA, USA) image-analysis software was used for healing assessment. Methods Twenty-seven traumatized single-rooted permanent teeth diagnosed with necrotic pulps and chronic periapical lesions were treated with non-surgical procedure using a tri-antibiotic paste and calcium hydroxide as intra-canal medication. Definitive obturation was performed with ProRoot MTA (Dentsply Tulsa Dental Specialties, Tulsa, OK, USA) or MTA+ Cerkamed (Cerkamed, Stalowa Wola, Poland). Control follow-ups were done three, six, 12, and 24 months following the completion of treatment. Results the positive clinical outcome was recorded in 24 (88.9%) cases, while radiographic success was present in 26 (96.3%) cases. A statistically significant decrease in the sizes of periapical lesions was consistently observed at follow-up periods (p  lt  0.001). There was no statistically significant difference between the two tested MTA materials (p > 0.05). Conclusion the MTA products were effective for the root canal obturation and appeared to contribute to the significant reduction or complete regression of periapical lesions in teeth treated. The presented procedure may be proposed for everyday clinical practice.
AB  - Uvod/Cilj Svrha ove studije bila je procena efikasnosti endodontskog tretmana traumatizovanih stalnih zuba sa nekrotičnom pulpom i hroničnim periapikalnim lezijama posle definitivne opturacije mineralnim trioksidnim agregatom (MTA). Softver za analizu slike Adobe Photoshop CS korišćen je za procenu regeneracije. Antibacterial effects of new endodontic materials based on calcium silicates

AB  - Background/Aim. The main task of endodontic treatment is to eliminate pathologically altered tissue, to disinfect root canal space and to obtain its three-dimensional hermetic obturation. The main purpose of this study was to evaluate antimicrobial activity of new endodontic nano-structured highly active calcium silicates based materials albo-mineral plyoxide carbonate aggregate (ALBO-MPCA) and calcium silicates (CS) in comparison to mineral trioxide aggregate (MTA +) and UltraCal XS (CH). Methods. The antimicrobial activity of materials was tested against Staphylococcus aureus (ATCC 25923) and Entero-coccus faecalis (ATCC 14506) strains, and following clinical isolates: Rothia dentocariosa, Enterococcus faecalis, Staphylococcus aureus, Streptococcus anginosus and Streptococcus vestibularis using a double layer agar diffusion test. The pH measurements were performed using the pH meter. Total amount of released ions was determined by inductively coupled plasma optical emission spectrometry (ICP-OES). Results. All tested materials showed the best antibacterial potential after 1 h of incubation. After 3h and 24h of the incubation period, the an-tibacterial potential of all tested materials were similar. The Agar diffusion test showed that ALBO-MPCA, CS and MTA + had similar inhibition zones (p > 0.05), except in the activity against Staphylococcus aureus where ALBO-MPCA showed better antimicrobial properties than MTA + in 3h and 24h of the incubation period (p  lt  0.05). Following 24h of the incubation, the inhibition zones were the strongest with CH against Staphylococcus aureus (16.67 ± 2.34 mm) followed by ALBO-MPCA (14.67 ± 1.21 mm) and the weakest with CS against Enterococcus faecalis (6.50 ± 1.76 mm). CH showed the highest pH, followed by ALBO-MPCA, CS and MTA +. Conclusion. The expressed antibacterial effects indicate that materials based on nano-structured highly active calcium silicates represent effective therapeutic agents for root canal obturation in one-visit apexification treatment, therefore they are recommend for further examination and clinical trials as they are proposed for MTA substitution.
AB  - Uvod/Cilj. Osnovni cilj endodonskog lečenja je eliminacija patološki izmenjenog tkiva, eliminacija infekcije korensko kanala i njegovo hermetičko trodimenzionalno zatvaranje. Cilj istraživanja je bio da se proceni antibakterijska aktivnost novih endodontskih nano-strukturiranih materijala na bazi visoko aktivnih kalcijum silikata albo-mineral polyoxide carbonate aggregate (ALBO-MPCA) i calcium silicates (CS) u odnosu na mineral trioxide aggregate (MTA +) i UltraCal XS (CH). Metode. Testirana je antibakterijska aktivnost materijala protiv Staphylococcus aureus (ATCC 25923) i Enterococcus faecalis (ATCC 14506), kao i kliničkih izolata: Rothia dentocariosa, Enterococcus faecalis, Staphylococcus aureus, Streptococcus anginosus i Streptococcus vestibularis pomoću agar difuzionog testa. Merenja pH vred-nosti obavljena su korišćenjem pH metra. Ukupan iznos oslobođenih jona određivan je pomoću ICP-OES. Rezultati. Svi testirani materijali pokazali su najbolji antibakterijski efekat nakon 1 h od inkubacije. Nakon 3 h i 24 h od inkubacije, antibakterijski efekat svih testiranih materijala bio je sličan. Agar difuzioni test pokazao je da materijali ALBOMPCA, CS i MTA+ ispoljavaju slične zone inhibicije rasta (p > 0.05) osim u slučaju Staphylococcus aureus, gde je materijal ALBO-MPCA pokazao bolje antibakterijsko dejstvo nego MTA+ nakon 3 h i 24 h od inkubacije (p  lt  0.05). Nakon 24 h od inkubacije, zone inhibicije su bile najizraženije u slučaju materijala CS protiv Staphylococcus aureus (16.67 ± 2.34 mm), zatim ALBO-MPCA (14.67 ± 1.21 mm), a najslabije u slučaju CS protiv Enterococcus faecalis (6.50±1.76 mm). Materijal CH ispoljio je najveću pH vrednost, zatim ALBOMPCA, CS i MTA+. Zaključak. Ispoljeni antibakterijski efekti ukazuju na to da materijali na bazi visoko aktivnih kalcijum silikata mogu da predstavljaju efikasnu zamenu za MTA u terapiji zuba sa nezavršenim rastom korena u jednoj poseti, te se stoga preporučuju za dalja klinička ispitivanja

Cilj istraživanja je bio da se proceni antibakterijska aktivnost novih endodontskih nano-strukturiranih materijala na bazi visoko aktivnih kalcijum silikata albo-mineral polyoxide carbonate aggregate (ALBO-MPCA) i calcium silicates (CS) u odnosu na mineral trioxide aggregate (MTA +) i UltraCal XS (CH). Metode. Testirana je antibakterijska aktivnost materijala protiv Staphylococcus aureus (ATCC 25923) i Enterococcus faecalis (ATCC 14506), kao i kliničkih izolata: Rothia dentocariosa, Enterococcus faecalis, Staphylococcus aureus, Streptococcus anginosus i Streptococcus vestibularis pomoću agar difuzionog testa. Merenja pH vred-nosti obavljena su korišćenjem pH metra. Ukupan iznos oslobođenih jona određivan je pomoću ICP-OES. Rezultati. Svi testirani materijali pokazali su najbolji antibakterijski efekat nakon 1 h od inkubacije. Nakon 3 h i 24 h od inkubacije, antibakterijski efekat svih testiranih materijala bio je sličan. Agar difuzioni test pokazao je da materijali ALBOMPCA, CS i MTA+ ispoljavaju slične zone inhibicije rasta (p > 0.05) osim u slučaju Staphylococcus aureus, gde je materijal ALBO-MPCA pokazao bolje antibakterijsko dejstvo nego MTA+ nakon 3 h i 24 h od inkubacije (p  lt  0.05). Nakon 24 h od inkubacije, zone inhibicije su bile najizraženije u slučaju materijala CS protiv Staphylococcus aureus (16.67 ± 2.34 mm), zatim ALBO-MPCA (14.67 ± 1.21 mm), a najslabije u slučaju CS protiv Enterococcus faecalis (6.50±1.76 mm). Materijal CH ispoljio je najveću pH vrednost, zatim ALBOMPCA, CS i MTA+. Zaključak. Ispoljeni antibakterijski efekti ukazuju na to da materijali na bazi visoko aktivnih kalcijum silikata mogu da predstavljaju efikasnu zamenu za MTA u terapiji zuba sa nezavršenim rastom korena u jednoj poseti, te se stoga preporučuju za dalja klinička ispitivanja",

Evaluation of toothbrush contamination

The aim of this paper was to evaluate the oral health, oral hygiene awareness and assess the degree of contamination of toothbrushes among students attending Secondary Medical School. Material and Methods: Sixty students (32 boys, 28 girls; mean age ≅ 15,7±2,1) attending highschool were randomly selected for this study. Each student included in the study filled out a questionnaire regarding his/her life habits and oral hygiene. Clinical examinations were initiated in order to determine the DMFT, as well as the CIP, CIT, CIA and CPITN indexes, based of which the assessment of oral health status was performed. One stack of fiber was collected from each toothbrush used by the participantes in the study, and than prepared for further microbiological sampling. Results: Only 11% of the students had the awareness of potential sources of toothbrushes contamination. The average value of DMFT was 3,2 (%D = 22,5; %M = 4,96; %F = 72,5). A statistically significant difference in comparison of the CPITN index and subjects' response to the professional plague removal in the last 12 months was identified (χ2 = 13,55; p= 0,033). Staphylococcus aureus, Streptococcus mutans, Micrococcus species and Streptococcus salivarius were most commonly present microorganisms. In most cases, G-positive bacilli or cocci were isolated, while the presence of Candidae albicans was identified in four samples. Conclusions: Raising the awareness of dental hygiene through the oral health education may improve better plaque control and subsequently the oral health. Hands-on training how to maintain the oral hygiene are not expensive and more over they are easy to be organized can be useful in oral heath promotion.

How to maintain oral health in children with respiratory diseases: Literature review

AB  - The most frequent chronic respiratory problems in childhood are asthma and cystic fibrosis (CF). The purpose of this paper is to review basic knowledge and recent advances in oral health and associated dental morbidities in children with asthma and CF. This review considered clinical trials and systematic reviews related to oral health in children with CRD. An online base Medline was searched to determine relevant papers, using the combination of the following terms: 'asthma', 'cystic fibrosis', 'caries', 'dental erosion', and 'oral health'. Oral health problems in children with chronic respiratory diseases (CRD) may be influenced by natural course of the disease, pharmacotherapy (inhalation therapy with bronchodilators and inhaled corticosteroids in asthmatic patients, systemic antibiotics and pancreatic enzyme replacement therapy in CF patients), medication administration technique and nutritional habits. Children with CRD may have higher prevalence of oral diseases. Patients and their parents, but also general paediatricians and pulmonologists, should be aware of importance of good oral health. Dental practitioners should be more informed about risk factors and specificities of oral health in these patients. Preventive measures, early diagnosis and effective treatment strategies in children with CRD can reduce occurrence of oral diseases and improve patient's quality of life.

AB  - Background: All objects put into a child's mouth could be hazardous in terms of trauma and toxic substance exposure. The aims of this study were to evaluate morphological characteristics of the primary teeth bite marks inflicted on various materials and to assess material wear using experimental model. Methods: Bite marks were analyzed on five materials: rubber, plastic, foil, wood, and silicone. In order to mimic children mouthing behavior an experimental setup has been designed using primary teeth placed in dentures and children's equipment specimens. Results: Deciduous teeth make visible and recognizable traces when using physiological forces on all investigated materials. The most significant material loss was revealed in silicone samples, but it has been observed in all material groups, while mouthing with incisors using higher mastication forces were identified as significant predictors for material wear. There were no significant differences between type, species, and morphological-morphometric characteristics of the bite marks which are made by incisors, canines, and molars. Conclusions: In the range of physiological bite forces, deciduous teeth lead to wear of material from which toys are made while the analysis of bite marks in children equipment could give some information regarding the risk of trauma and exposure.

Characterization of glass ionomer cements stored in various solutions

AB  - The aim of this work was to evaluate the nano-mechanical properties of glass ionomer materials, the ion concentrations at the surfaces in relation to the storage media and the pH environments using a scanning electron microscope with an energy-dispersive spectrometer (SEM/EDX). the glass-ionomer-based materials, Fuji Triage (FT), Fuji VIII (FVIII). Fuji IX GP (FIX), were analyzed. The sample comprised 45 cured cement disks. Five specimens of each tested material were placed in 3 storage solutions (saline, acidic solution with pH of 5.5, NaF solution with 0.05 % of fluoride). Nano-indentations were performed with a force up to 30 mN, penetration depths of 2500-2700 nm for 1 d and 21 d after setting. The EDX evaluation was carried out for each experimental disk, identifying the ions: O, Al, Sr, Si, F,Na, P, Ca. The level of significance was placed at p  lt  0.05.The highest fluoride proportion at the specimen surface was recorded in the FT material. FT also exhibited the lowest fluoride ions content when stored in low-pH environments compared with the other tested materials (p  lt  0.05). The surface hardness of the tested materials decreased from 1.377 GPa (in saline) to 0.03 GPa (in acid). The Young's modulus varied from 14.35 GPa to 0.112 GPa, depending on the material type (Fuji VIII>Fuji IX>FT) (p  lt  0.001) and the storage media (p  lt  0.001). Both the mechanical and cariostatic surface properties of commercially available glass ionomer materials are affected by the storage media.

AB  - Purpose: The objective was to assess the effect of fissure depth and enamel surface preparation protocols on penetration of resin-based, giomer and glass-ionomer fissure sealants. Materials and Methods: Ninety extracted third molars were assigned into three groups according to the material used. All three groups were subsequently divided depending on tooth preparation: no pretreatment or surface preparation with 10% polyacrylic acid, 37% phosphoric acid, or self-etching adhesive. In addition, Er,Cr:YSGG laser or air abrasion were employed in all subgroups. The penetration was evaluated using scanning electron microscopy. Results: The groups that were pretreated with an appropriate acid resulted in superior penetration of glass-ionomer and resin-based sealants when compared with other surface preparation protocols (p  lt  0.05). The exception was teeth treated with both air abrasion and acid etching. Both preparation protocol and fissure depth significantly affected the sealant penetration (p  lt  0.05), but fissure depth had a greater impact on fissure penetration than did enamel surface pretreatment. Conclusion: All investigated materials exhibit similar properties regarding sealant penetration. Penetration of a fissure sealant is significantly influenced by the fissure depth. In regard to enamel surface preparation protocol, a pretreatment with phosphoric acid in resin-based and polyacrylic acid in glass-ionomers appears to be essential in obtaining the adequate penetration of a sealing material.

abstract = "Purpose: The objective was to assess the effect of fissure depth and enamel surface preparation protocols on penetration of resin-based, giomer and glass-ionomer fissure sealants. Materials and Methods: Ninety extracted third molars were assigned into three groups according to the material used. All three groups were subsequently divided depending on tooth preparation: no pretreatment or surface preparation with 10% polyacrylic acid, 37% phosphoric acid, or self-etching adhesive. In addition, Er,Cr:YSGG laser or air abrasion were employed in all subgroups. The penetration was evaluated using scanning electron microscopy. Results: The groups that were pretreated with an appropriate acid resulted in superior penetration of glass-ionomer and resin-based sealants when compared with other surface preparation protocols (p  lt  0.05). The exception was teeth treated with both air abrasion and acid etching. Both preparation protocol and fissure depth significantly affected the sealant penetration (p  lt  0.05), but fissure depth had a greater impact on fissure penetration than did enamel surface pretreatment. Conclusion: All investigated materials exhibit similar properties regarding sealant penetration. Penetration of a fissure sealant is significantly influenced by the fissure depth. In regard to enamel surface preparation protocol, a pretreatment with phosphoric acid in resin-based and polyacrylic acid in glass-ionomers appears to be essential in obtaining the adequate penetration of a sealing material.",

Stem cells in tissue engineering: Dynamic cultivation requirement

AB  - Stem cells have shown great potential for in vitro tissue engineering, regenerative medicine, cell therapy and pharmaceutical applications. All these applications, especially in clinical trials, will require guided production of high-quality cells. Traditional culture techniques and applications have been performed for the majority of primary and established cell lines and standardized for various analyses. Still, these culture conditions are unable to mimic dynamic and specialized three-dimensional microenvironment of the stem cells' niche from in vivo conditions. In an attempt to provide biomimetic microenvironments for stem cells in vitro growth, three-dimensional culture techniques have been developed. In our study advantages of newly developed porous scaffolds as the most promising in vitro imitation of niche that provides physical support, enables cell growth, regeneration and neovascularization, while they are replaced in time with newly created tissue was explained. Furthermore, dynamic cultivation techniques have been described, as new way of cell culturing that will be the main subject of our future research. In that manner, by developing an optimal dynamic culturing method, high-quality new cells and tissues would be possible to obtain, for any future clinical application.
AB  - Matične ćelije su pokazale veliki potencijal za primenu u tkivnom inženjerstvu u in vitro uslovima, regenerativnoj medicini, lečenju matičnim ćelijama i farmaceutskoj primeni. Sve ove grane, posebno primena u kliničkim istraživanjima, zahtevaće kontrolisano stvaranje visokokvalitetnih ćelija. Tradicionalnim tehnikama izolovana je većina primarnih ćelijskih kultura i ćelijskih linija, i vremenom su one postale standardne tehnike za različite ćelijske analize. Ipak, ovi uslovi gajenja nisu u mogućnosti da imitiraju dinamične, trodimenzionalne uslove mikrosredine niše matičnih ćelija iz in vivo uslova. U pokušaju da se obezbede navedeni uslovi i u in vitro uzgajanju ćelija, razvile su se trodimenzionalne tehnike gajenja ćelija. U ovom preglednom radu opisujemo prednosti novorazvijenih poroznih nosača ćelija, kao ključnih činilaca u imitaciji ćelijske niše koji obezbeđuju mehaničku potporu, omogućuju rast ćelija, regeneraciju i razvoj novih krvnih sudova, dok vremenom bivaju razgrađeni i zamenjeni novostvorenim tkivom. Dalje, tehnike dinamičnog gajenja ćelija su opisane kao vid novog načina gajenja ćelijskih kultura koji predstavlja i pravac naših budućih istraživanja. U tom smislu, razvijajući optimalan, dinamički model gajenja ćelija, biće moguće obezbediti nove ćelije i tkiva visokog kvaliteta za sva dalja klinička istraživanja.

Nanostructured endodontic materials mixed with different radiocontrast agentsbiocompatibility study

AB  - The aim of this study was to investigate the biocompatibility of nanostructured materials based on highly active calcium silicates mixed with different radiocontrast agents in comparison to MTA(+) using in vitro and in vivo model. Morphology of materials' samples was analyzed using SEM while the phase compositions were identified by XRD. pH values of materials' suspensions were conducted by pH-meter. The cytotoxicity of materials' solutions was tested by MTT test (100, 50, 25 and 12.5mg/ml). LDH and H-3-thymidine assay were utilized for biocompatibility investigations of materials' eluates (24h, 7 day and 21 day). Eighteen Guinea pigs were used for intramuscular implantation, as teflon tubes with freshly prepared materials were placed into intramuscular pockets. All samples were composed of round and needle-like particles equally distributed with Ca/Si ratio 2.7 at%, with the presence of hydrated calcium silicate phases. The pH values of ALBO-MPCA(1) and ALBO-MPCA(2) were high alkaline, while in case of MTA(+) they were lower and continuously declined (p lt 0.05). Investigated materials didn't exhibit dose-dependent effect on metabolic activity of L929 cells (p>0.05). Significant differences in the percentage of cytotoxicity between diluted and undiluted extracts between all tested materials after 24h and 7 day were noticed (p lt 0.05). Increase in L929 cells proliferation was noticed in case of undiluted eluates of ALBO-MPCA(1) and ALBO-MPCA(2) after 7 day (p lt 0.05). There were no statistically significant differences in the intensity of inflammatory response between investigated materials and control group after 60 day (p>0.05). Evaluation of biocompatibility of both ALBO-MPCA(1) and ALBO-MPCA(2) indicate their potential clinical use. [GRAPHICS] .

Glass-ionomer fissure sealants: Clinical observations up to 13 years

AB  - Objectives: The aim of this study was to clinically examine the retention and caries-preventive effect of a glass-ionomer material for fissure sealing in newly erupted permanent molars. Methods: A total of 1736 newly erupted permanent molars were sealed in 480 children aged 5-15 years. Sealants were evaluated yearly and recorded as "completely retained", "partially retained", or "completely lost". Teeth were also evaluated for presence of caries. Results: Sealants were observed for 2-13 years. The mean follow-up time was 5 years. After one year, 69% of the sealants were fully retained and 21% were partially retained. Thereafter, the cumulative retention rate decreased. Starting from 8th year of follow-up, no "completely retained" sealant could have been observed. At the end of the 13-year follow-up period, 76% of sealants were "completely lost". On the other hand, 99% and 65% of observed occlusal surfaces were caries-free after one and 13 years, respectively. During the observation period, 62 teeth developed caries in surfaces other than occlusal. Survival rates were significantly better for second permanent molars than for first permanent molars (p  lt  0.001). No influence of caries risk on caries incidence has been noted. Conclusions: Although the retention rate of the glass-ionomer material for fissure sealing was low, it appears to have prevented dental caries in 65% of newly erupted permanent molars evaluated after thirteen years of placement. Clinical significance: The present study reports long-term clinical performance of glass-ionomer fissure sealants in patients with low, moderate and high caries-risk. The study shows that caries risk does not influence survival of glass-ionomer sealants, which proved to be beneficial for long-term caries protection.

AB  - This study examined the potential of a new porous calcium hydroxyapatite scaffold covered with poly (lactide-co-glycolide) (PLGA) as a bone substitute, identifying its advantages over Geistlich Bio-Oss (R), considered the gold standard, in in vivo biofunctionality investigations. Structural and morphological properties of the new scaffold were analyzed by scanning electron and atomic force microscopy. The biofunctionality assays were performed on New Zealand white rabbits using new scaffold for filling full-thickness defects of critical size. The evaluated parameters were: the presence of macrophages, giant cells, monoocytes, plasma cells, granulocytes, neo-angiogenesis, fibroplasia, and the percentage of mineralization. Parallel biofunctionality assays were performed using Geistlich Bio-Oss (R). The appearance of bone defects 12 weeks after the new scaffold implantation showed the presence of a small number of typical immune response cells. Furthermore, significantly reduced number of capillary buds, low intensity of fibroplasia and high degree of mineralization in a lamellar pattern indicated that the inflammation process has been almost completely overcome and that the new bone formed was in the final phase of remodeling. All biofunctionality assays proved the new scaffold's suitability as a bone substitute for applications in maxillofacial surgery. It showed numerous biological advantages over Geistlich Bio-Oss (R) which was reflected mainly as a lower number of giant cells surrounding implanted material and higher degree of mineralization in new formed bone.

The appearance of bone defects 12 weeks after the new scaffold implantation showed the presence of a small number of typical immune response cells. Furthermore, significantly reduced number of capillary buds, low intensity of fibroplasia and high degree of mineralization in a lamellar pattern indicated that the inflammation process has been almost completely overcome and that the new bone formed was in the final phase of remodeling. All biofunctionality assays proved the new scaffold's suitability as a bone substitute for applications in maxillofacial surgery. It showed numerous biological advantages over Geistlich Bio-Oss (R) which was reflected mainly as a lower number of giant cells surrounding implanted material and higher degree of mineralization in new formed bone.",

AB  - The aim of this study was to analyze and emphasize the importance of contemporary pharmacy which enabled development of modern health care system in Jagodina district in Serbia. Material and method: The research was descriptive, document analysis methods, desk analysis of secondary data and of historical synthesis were used. It covered the historical period from the 12th century till the modern age. The first owner of the pharmacy in Serbia in XIX century was Anton Delini in 1813. The first pharmacy in Jagodina was founded by Mr. Djordje Krstic on August 22nd, 1852 and it was very important for the health care development in Jagodina district. Conclusions: Pioneers in the development of pharmacy in Jagodina had an important impact on the socio-economic and cultural development of this region.
AB  - Cilj istraživanja je analiza i isticanje početka razvoja modernog apotekarstva koje je uticalo na razvoj zdravstvene svesti u Jagodini. Materijal i metod: istraživanje je po tipu opisno, korišćene su metode dokumentacione analize, desk analiza sekundarnih podataka i sinteza istorijskih činjenica. Posmatrani istorijski period je od XII veka do savremenog doba. Rezultati: Vlasnik prve apoteke u Srbiji u 19. veku (1813) bio je Anton Delini. Apotekar Đorđe Krstić je 22. avgusta 1852. godine otvorio prvu apoteku u Jagodini. Zaključak: Otvaranje prvih apoteka predstavljao je važan korak u oblasti razvoja zdravstvene kulture u Pomoravskom okrugu. Na taj način je omogućen razvoj farmacije i savremene medicine. Pioniri apotekarstva su ostavili veliki trag na socijalno-ekonomski i kulturološki razvoj sredine u kojoj su živeli i radili.

AB  - BACKGROUND: Dentists have a high prevalence of musculoskeletal (MS) pain, which is the most common symptom associated with work-related musculoskeletal disorders (WMSDs). To overcome this problem, identification of the risk factors and preventive measures for MS pain are of paramount importance to dentists in order to improve their quality of life and work. OBJECTIVES: The aims of this study were to recognize the risk factors for MS pain and their impact on dental work, as well as to identify preventive measures of MS pain among dentists. METHODS: Self-reporting questionnaire consisting of 78 questions was exclusively developed for the study and sent to 500 working active dentists in Serbia. RESULTS: Response rate was 71.2% (356 dentists). The prevalence of MS pain was 82.6% among dentists. The main risk factors for MS pain were advanced age, female dentists, presence of chronic diseases, long working hours, and high frequency of treated patients. The most effective preventive measures in preventing MS pain were massage treatments and physical activities. Followed by use of ergonomically designed equipment, correct and dynamic working positions, and an adequate workflow organization. CONCLUSION: The risk factors for MS pain and their impact on dental work should widely be disseminated among dentists. Importantly, proper implementation in everyday life of adequate preventive measures is essential for preventing MS pain and development of WMSDs.

AB  - Bioactive potential of apigenin derivative apigenin-7-O-glucoside related to its antifungal activity on Candida spp. and cytotoxic effect on colon cancer cells was studied and compared with bioactive potential of apigenin. Antifungal activity was tested on 14 different isolates of Candida spp. using membrane permeability assay, measuring inhibition of reactive oxidative species and inhibition of CYP51 C. albicans enzyme. Cytotoxic potential of apigenin- 7-O-glucoside was tested on colon cancer HCT116 cells by measuring cell viability, apoptosis rate and apoptosis- and colon cancer-related gene expression. Obtained results indicated considerable antifungal activity of apigenin-7-O-glucoside towards all Candida isolates. Breakdown of C. albicans plasma membrane was achieved upon treatment with apigenin-7-O-glucoside for shorter period of time then with apigenin. Reduction of intra-and extracellular reactive oxidative species was achieved with minimum inhibitory concentrations of both compounds, suggesting that reactive oxidative species inhibition could be a mechanism of antifungal action. None of the compounds exhibited binding affinity to C. albicans CYP51 protein. Besides, apigenin-7-O-glucoside was more effective compared to apigenin in reduction of cell's viability and induction of cell death of HCT116 cells. Treatment with both compounds resulted in chromatin condensation, apoptotic bodies formation and apoptotic genes expression in HCT116 cells, but the apigenin-7-O-glucoside required a lower concentration to achieve the same effect. Compounds apigenin-7-O-glucoside and apigenin displayed prominent antifungal potential and cytotoxic effect on HCT116 cells. However, our results showed that apigenin-7-O-glucoside has more potent activity compared to apigenin in all assays that we used.

AB  - Introduction: The aim of this article was to analyze biocompatibility and bioactivity of new endodontic materials on the basis of nanosynthesized calcium silicates (ALBO-MPCA(1) and ALBO-MPCA(2)) combined with different radiopacifiers in comparison with MTA(+). Methods: Morphology of the samples was studied by scanning electron microscopy, and the pH and ion release analysis were also assessed. Biocompatibility of materials' eluates (24-hour, 7-day, and 21-day) was conducted by using MIT test. Twelve New Zealand white rabbits were used for intraosseous implantation. Four calvarial defects per animal were created and filled with freshly prepared investigated materials. Results: Samples mostly consisted of agglomerates built up from nanoparticles, preferably spherical and rod-like. There was no significant difference among pH values of materials' eluates after 24 hours (P>.05). The amount of calcium and aluminum ion release decreased, whereas the amount of magnesium and bismuth (ALBO-MPCAl, MTA(+)) and barium (ALBO-MPCA(2)) increased during 21-day period. The metabolic activity of cells increased after the extraction time, except in case of undiluted elutes of ALBO-MPCA(2) and ALBO-MPCAI (21-day). Histologic analysis of the samples revealed newly formed bone tissue with moderate inflammation for all investigated materials, which subsided during 90-day period to mild. Both MIA(+) and ALBO-MPCAI were in direct contact with the newly formed bone tissue. After 90 days, statistically significant difference in hard tissue formation was observed in comparison of MIA(+) and ALBO-MPCki with control group (P  lt .05). Conclusions: Experimental materials ALBOMPCA, and ALBO-MPCA(2) possess both biocompatibility and bioactivity. Because ALBO-MPCAA provokes favorable biological response, it is especially good candidate for further clinical investigations.

Relationship between IL-1β production and endodontic status of human periapical lesions

AB  - Background/Aim. Apical periodontitis is mainly caused by bacterial infection within the root canal and periapical bone destruction which are prominent features of this lesion. The aim of this study was to determine the quantity of interleukin-1β in the tissues of periapical lesions and to analyze its relationships with: lesion size, previous treatments and pathohistological finding of involved teeth. Methods. Periapical tissues were obtained from patients undergoing periapical surgery. Out of all 80 cases included in the study, 24 had no previous endodontic treatment (open lesions), 37 were with endodontic failure (closed lesion) and in 15 cases root canal retreatment was performed few months before the surgery. By excluding four samples, the total of 76 samples, consisted of periapical lesions and the apical part of the tooth root, was collected. Each periapical tissue sample was divided into two equal parts. The one half of each lesion was used for quantification of interleukin-1β in tissue homogenates by the enzyme-linked immunosorbent assay (ELISA) method. The other part of each lesion was used for histopathological evaluation. Results. For each of the tissue homogenates, the quantity of interleukin-1β was measured, and it ranged from 0.6 pg/mg up to 74 pg/mg. There was no significant difference between the symptomatology and amount of interleukin-1β. Statistical data analysis showed a moderate correlation between lesion size and interleukin-1β measured values. The highest levels of interleukin-1β corresponded with chronic lesions in the stages of acute exacerbation and granulomas in early developing stages. Persistant granulomas, scar tissues, non-inflamed cysts and teeth with recently finished endodontic treatments showed a significantly lower level of interleukin-1β. Conclusion. The study results suggest that the differences in quantity of interleukin-1β correlate to lesion progression and phases of development.
AB  - Uvod/Cilj. Periapikalni periodontitis najčešće je prouzrokovan bakterijskom infekcijom unetom putem kanala zubnog korena i periapikalnom destrukcijom kosti koje predstavlja glavno obeležje ovih lezija. Cilj ovog istraživanja bio je da se odredi količina interleukina-1β u uzorcima periapikalnih lezija i da se analizira njihova povezanost sa: veličinom lezija, postojanjem prethodnih endodontskih tretmana i patohistološkim nalazima. Metode. Ispitivani su uzorci periapikalnih lezija sakupljeni tokom periapikalnih operativnih intervencija. Od svih 80 bolesnika uključenih u studiju, 24 su pripadali bolesnicima koji nisu podvrgnuti endodontskom tretmanu pre apikotomije (otvorene lezije), 37 su tretirani kao endodontski neuspeh (zatvorene lezije), a kod 15 bolesnika endodontski retreatman je sproveden nekoliko meseci pre oralno-hirurške intervencije; četiri uzorka su isključena iz studije. Kod starih granuloma, ožiljačnih tkiva, zuba sa nedavno završenim endodontskim retretmanom i neinflamiranih cista, vrednosti interleukina-1β bile su značajno niže. Zaključak. Rezultati istraživanja pokazuju da su razlike u količini interleukina-1β u korelaciji sa progresijom lezija i fazama njihovog razvoja.

AB  - Background/Aim: Lately, fully innovative sol-gel method with high-temperature self-propagating reaction was used for the synthesis of new nanostructured endodontic materials, in combination with different radiopacifiers: bismuth (ALBO-MPCA1) and barium (ALBO-MPCA2). The aim of this study was to investigate the biocompatibility of nanostructured endodontic materials based on highly active calcium silicates and mixed with different radiopacifiers in comparison to MTA+ using human stem cells from the apical papilla- SCAP cells. Material and Methods: Morphology of the samples was studied by SEM. The tested materials were mixed with distilled water in a ratio 2:1 (m/m). Fifteen minutes after the preparation, samples were used in the experiment. The biocompatibility of fresh materials, after 3h and 7 days, was tested using 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide- MTT test. Results: Samples mostly consisted of spherical and rode-like. The relative viability of cells increased following the exposure time. Conclusion: The biocompatibility of synthesized materials is comparable to the control material MTA+, and therefore these materials can be recommended for further clinical studies.

Diode laser irradiation in endodontic therapy through cycles: In vitro study

AB  - Background/Aim: The aim of this in vitro study was to investigate the influence of irradiation cycles and resting periods, on thermal effects on the external root surface during root canal irradiation of two diode laser systems (940 nm and 975 nm), at output powers of 1 W and 2 W in continuous mode. In previous studies the rising of temperature above 7°C has been reported as biologically accepted to avoid periodontal damage on the external root surface. Material and Methods: Twenty human inferior incisors were randomly distributed into four groups, the 940 nm, and the 975 nm diode laser irradiation, both with an output power of 1 W and 2 W, in continuous mode. The thermographic camera was used to detect temperature variations on the external root surface. Digital radiography of the samples was made. Results: After three cycles of irradiation, at apical third of the root, mean temperature variation by 940 nm diode laser irradiation was 2.88°C for output power of 1 W, and 6.52°C for output power of 2 W. The 975 nm laser caused a higher temperature increase in the apical region, with temperature variation of 13.56°C by an output power of 1 W, and 30.60°C at 2 W, with a statistical significance of p ≤ 0.0001 between two laser systems compared for the same power. The resting periods of 20 s between cycles were enough to lower temperature under 7°C in the case of 1 W and 2 W for 940 nm diode laser, while for 975 nm laser, after three irradiation cycles overheating occurred at both output power rates. Conclusion: Three cycles irradiation of 940 nm diode laser, with resting periods of 20 seconds, allowed safe usage of 1 W and 2 W in CW for endodontic treatment. For 975 nm at a power rate of 1 W, the last resting period drop the temperature near the safe limit and it came under 7°C in a period less than a minute, while at the power of 2 W the resting periods were not long enough for the safe temperature decrease.

AB  - Background/Aim. The longevity of a dental restoration may be predicted to some degree by its adhesive ability, and this, in turn, can be measured by bond strength testing between restorative materials and tooth structure. The aim of this study was to test an innovative joggle lap shearing jig that integrates the tooth and the entire biomechanical unit into testing, to compare the shear bond strengths of Class I occlusal composite restorations in deep cavity preparations lined with Dycal, Dycal LC, conventional glass ionomer or resin-modified glass ionomer. The mode of failure (adhesive, cohesive, mixed) after debonding was determined by stereomicroscopy. Methods. A total of 150 standardized occlusal cavities were prepared and divided into five groups. The group I cavities (n = 30) were coated with adhesive (ExciTE®F) and filled directly with composite (TetricEvoCeram). The group II and III cavities were lined with Dycal (n = 30) or Dycal LC (n = 30) before placing composite. The groups IV and V specimens were based with Fuji IX (n = 30) or Fuji II LC (n = 30). Shear bond strengths were determined with a universal testing machine and fractured bonding sites were analyzed under stereomicroscope. The mean bond strengths were analyzed using one-way ANOVA test (p  lt  0.05) and the means between the groups were analyzed with Student's t-test. Results. The shear bond strength (MPa) of composite restorations in cavities without base (23.91 ± 4.54) was higher than cavities lined with Fuji II LC (17.45 ± 2.74), Fuji IX (8.76 ± 2.57), Dycal LC (13.07 ± 1.84) or Dycal (6.12 ± 1.28). The results using the jogged lap shearing jig were consistent with the literature. Conclusion. The shear bond strength of occlusal composite restorations in deep cavities without liners was greater than cavities lined with Fuji II LC > Fuji IX > Dycal LC > Dycal.
AB  - Uvod/Cilj. Trajnost zubnih nadoknada može se donekle predvideti vstom adhezivne sposobnosti materijala i može se meriti testiranjem snage adhezije restorativnih materijala i zubnih struktura. Cilj ove studije bio je da se da se testira preklapanje spoja koji povezuje zub i biomehaničku jedinicu i da se uporedi jačina veze okluzalnih kompozitnih ispuna postavljenih u duboke kavitete preko podloge od Dycal, Dycal LC, konvencionalnog ili smolom-modifikovanog glas-jonomer cementa. Metode. Ukupno 150 standardizovanih okluzalnih kaviteta bilo je podeljeno u pet grupa (n = 30): I - kaviteti premazani adhezivom (ExciTE®F) i direktno ispunjeni kompozitom (TetricEvoCeram); II i III - kaviteti sa podlogom od Dycal ili Dycal LC pre postavljanja kompozitnog materijala; IV i V - uzorci sa bazom od Fuji IX ili Fuji II LC (n = 30). Jačina vezivne snage određena je pomoću univerzalne mašine, a način neuspeha (adhezivna, kohezivna, mešovita fraktura) određen je stereomikroskopom. Srednje vrednosti su analizirane pomoću ANOVA testa (p  lt  0,05), a značajnost razlika između grupa analizirana je Student-ovim t-testom. Rezultati. Jačina vezivne snage (MPa) kompozitnih ispuna u kavitetima bez podloge (23,91 ± 4,54) bila je veća u poređenju sa ispunima postavljenim preko Fuji II LC (17,45 ± 2,74), Fuji IX (8,76 ± 2,57), Dycal LC (13,07 ± 1,84) ili Dycal (6.12 ± 1.28). Zaključak. Smicanje ili pomaknuće okluzalnih kompozitnih ispuna u dubokim kavitetima bez lajnera je veće nego u kavitetima postavljenim preko Fuji II LC > Fuji IX > Dycal LC > Dycal.

The longevity of a dental restoration may be predicted to some degree by its adhesive ability, and this, in turn, can be measured by bond strength testing between restorative materials and tooth structure. The aim of this study was to test an innovative joggle lap shearing jig that integrates the tooth and the entire biomechanical unit into testing, to compare the shear bond strengths of Class I occlusal composite restorations in deep cavity preparations lined with Dycal, Dycal LC, conventional glass ionomer or resin-modified glass ionomer. The mode of failure (adhesive, cohesive, mixed) after debonding was determined by stereomicroscopy. Methods. A total of 150 standardized occlusal cavities were prepared and divided into five groups. The group I cavities (n = 30) were coated with adhesive (ExciTE®F) and filled directly with composite (TetricEvoCeram). The group II and III cavities were lined with Dycal (n = 30) or Dycal LC (n = 30) before placing composite. The groups IV and V specimens were based with Fuji IX (n = 30) or Fuji II LC (n = 30). Shear bond strengths were determined with a universal testing machine and fractured bonding sites were analyzed under stereomicroscope. The mean bond strengths were analyzed using one-way ANOVA test (p  lt  0.05) and the means between the groups were analyzed with Student's t-test. Results. The shear bond strength (MPa) of composite restorations in cavities without base (23.91 ± 4.54) was higher than cavities lined with Fuji II LC (17.45 ± 2.74), Fuji IX (8.76 ± 2.57), Dycal LC (13.07 ± 1.84) or Dycal (6.12 ± 1.28). The results using the jogged lap shearing jig were consistent with the literature. Conclusion. The shear bond strength of occlusal composite restorations in deep cavities without liners was greater than cavities lined with Fuji II LC > Fuji IX > Dycal LC > Dycal., Uvod/Cilj. Trajnost zubnih nadoknada može se donekle predvideti vstom adhezivne sposobnosti materijala i može se meriti testiranjem snage adhezije restorativnih materijala i zubnih struktura. Antimicrobial synergism and cytotoxic properties of Citrus limon L., Piper nigrum L. and Melaleuca alternifolia (Maiden and Betche) Cheel essential oils

AB  - Objectives The chemical composition, antimicrobial and synergistic effect, and cytotoxic activity of Citrus limon (lemon), Piper nigrum (green pepper) and Melaleuca alternifoila (tea tree) essential oils (EOs) were investigated. Methods Chemical analyses of essential oils were tested by GC-FID and GC-MS spectroscopy. The antimicrobial activity assay was conducted using microdilution method against several oral bacteria and Candida spp. originating from the humans with oral disorders. The synergistic antimicrobial activity was evaluated using checkerboard method. The cytotoxicity evaluation of EOs was assessed using MTT test. Key findings Limonene (37.5%) and beta-pinene (17.9%) were the major compounds in C. limon oil, -pinene (34.4%), delta-3-carene (19.7%), limonene (18.7%) and alpha-pinene (10.4%) in P. nigrum oil and terpinen-4-ol (38.6%) and gamma-terpinene (21.7%) in M. alternifolia oil. The broad-spectrum antimicrobial activity was achieved by tested three EOs, with C. limon oil being the strongest against bacteria and M. alternifolia oil strongest against fungi. The EOs demonstrated synergism; their combined application revealed an increase in antimicrobial activity. All tested essential oils showed lower cytotoxic activity in comparison with the positive control, and the obtained results confirmed a dose-dependent activity. Conclusions The results of this study encourage use of tested EOs in development of a novel agent intended for prevention or therapy of corresponding oral disorders.
