AB  - Introduction
This study aimed to perform a more precise estimation of the association between tumor necrosis factor-alpha (TNF-α) – 308 G/A single nucleotide polymorphism (SNP) and the risk of development of AP and its phenotypes based on all available published studies.
Methods
The study was carried out according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines and is registered in PROSPERO (CRD42020176190). The literature search was conducted via: Clarivate Analytics’ Scopus, PubMed, Cochrane Central Register of Controlled Trials and China National Knowledge Infrastructure databases, from inception to December 2020 with no language restrictions. Two reviewers were involved independently in study selection, data extraction and appraising the studies that were included. The quality of included studies was evaluated using the Strengthening the Reporting of Genetic Association (STREGA) and the Grading of Recommendations Assessment, Development and Evaluation (GRADE) system. Frequencies of genotypes and alleles of TNF-alpha (G>A 308, rs1800629) gene, with 95% Odds ratio was used.
Results
Four studies met the inclusion criteria with moderate risk of bias. This study revealed no significant association between TNF-α – 308 G/A SNP and AP, and the risk of AP development. Moreover, there was no significant association between genotype or allele frequency distribution and clinical manifestations (acute versus chronic) of AP. The certainty of evidence per GRADE was very low.
Conclusions
Due to very low certainty of evidence, whether there is an association between TNF-α – 308 G/A SNP and AP, warrants further well-designed multi-centric studies to adjudicate a better understanding of the role of genetic factors in the etiopathogenesis of AP.

 This study aimed to summarize data on apical periodontitis (AP) and nonsurgical root canal treatment (NSRCT) prevalence and risk factors related to age, gender, and quality of restorative and endodontic treatment in the general population from cross-sectional studies published between 2012 and 2020.
 
 Methods
 An electronic search was performed in the following databases: Web of Science, Scopus, and PubMed. The conducted literature search covered studies published between 2012 and 2020, without restrictions on language. The STROBE and NOS tools were used for quality assessment of the included studies.
 
 Results
 Sixteen articles were included in the review. In total, 200,041 teeth were examined. On average, 6.3% of teeth had AP, and 7.4% had NSRCT. Forty-one percent of RCT teeth had AP, and 3.5% of untreated teeth had AP. Female patients were less prone to AP in endodontically treated teeth only, compared with male patients (P < .001). Variable stratification of age subgroups among included studies prevented us from conducting a meta-analysis. An increase in AP frequency was found in teeth with inadequate restorative and endodontic treatment (P < .001 and P < .001, respectively). Because of high heterogeneity, these results should be taken with caution.
 
 Conclusions
 There is an increased AP prevalence in the adult general population compared with data from 2012 (6.3% versus 5.4%) in both endodontically treated (41.3% versus 35.9%) and untreated teeth (3.5% versus 2.1%). In addition, AP developed less frequently in female patients with endodontically treated teeth and in teeth with inadequate compared with adequate restorative and endodontic treatment.

AB  - Objectives: This study aimed to investigate whether Epstein-Barr virus (EBV) positive periapical lesions exhibited higher mRNA levels of Notch signalling molecules (Notch2 and Jagged1), bone resorption regulators (receptor activator of nuclear factor kappa-? ligand (RANKL) and osteoprotegerin (OPG)), and proinflammatory cytokines (tumour necrosis factor-alpha (TNF-?), interleukin-1 beta (IL-1?) and IL-6) compared to EBV negative lesions. Additionally, the potential correlation between investigated molecules in periapical lesions was analyzed. Materials and methods: Sixty-four apical periodontitis lesions were obtained subsequent to standard apicoectomy procedure. The presence of EBV was determined using nested PCR. Based on the presence of EBV all periapical lesions were divided into two groups, 29 EBV positive and 35 EBV negative lesions. A reverse transcriptase real-time PCR was used to determine mRNA levels of Notch2, Jagged1, RANKL, OPG, TNF-?, IL-1? and IL-6. Results: Significantly higher mRNA levels of Notch2, Jagged1, RANKL and IL-1? were observed in EBV positive compared to EBV negative lesions. Significant positive correlation was present between Notch2 and Jagged1, Jagged1 and RANKL, and IL-? and TNF-? in EBV positive periapical lesions. Conclusions: Notch signalling pathway may be involved in alveolar bone resorption in apical periodontitis lesions infected by EBV.

Association of polymorphisms in TNF-alpha, IL-1 beta, GSTM and GSTT genes with apical periodontitis: is there a link with herpesviral infection?

AB  - Aim To investigate the possible association between TNF alpha (-308 G/A) and IL-1 beta (-511 C/T) single nucleotide polymorphisms (SNPs) and GSTT and GSTM deletion polymorphisms and risk of apical periodontitis (AP) development, and determine the association of different genotypes with the presence of herpesviral infection in AP. Methodology The study included 120 periapical lesions and 200 control samples. Gene polymorphism analysis was performed using either polymerase chain reaction (PCR) or PCR/ restriction fragment length polymorphism (RFLP). Relative gene expression of TNF-alpha and IL-1 beta was analysed using reverse transcriptase - real-time PCR. The presence of Epstein-Barr virus (EBV) and human cytomegalovirus (HCMV) was assessed by nested PCR. Chi-square and Fisher's exact tests and logistic regression analyses were done for polymorphisms, whilst Mann-Whitney U-test was performed for the expression analysis. The expected frequency of variants was analysed by the Hardy-Weinberg equilibrium test. Results TNF-alpha (-308 G/A) SNP increased AP susceptibility for heterozygous (odds ratio (OR) = 1.72, 95% confidence interval (CI) = 1.06-2.80, P = 0.027) and homozygous (OR = 8.55, 95% CI = 1.77-41.36, P  lt  0.001) carriers of the variant A allele. On the other hand, IL-1 beta (-511 C/T) polymorphism exerted a protective effect both in heterozygotes (OR = 0.540, 95% CI = 0.332-0.880, P = 0.013) and homozygotes (OR = 0.114, 95% CI = 0.026-0.501, P  lt  0.001). In addition, GSTM1 and GSTT1 null genotypes separately, as well as concomitantly, were associated with an increased risk for AP development (P  lt  0.001). The null GSTT1 genotype increased approximately twice the risk of Epstein-Barr infection (EBV) in AP (OR = 2.17, 95% CI = 1-4.71, P = 0.048), whilst TNF-alpha SNP decreased it, both in heterozygotes (OR = 0.20, 95% CI = 0.08-0.48, P  lt  0.001) and AA homozygotes (OR = 0.07, 95% CI = 0.01-0.37, P = 0.001). Conclusions GSTM and GSTT deletion polymorphisms, as well as TNF alpha (-308 G/A) SNP, are associated with increased risk, whereas IL-1 beta (-511 C/T) polymorphism decreases the risk of AP development. GSTT and TNF alpha polymorphisms also appear to modulate the risk of EBV infection in Serbian patients with AP.

AB  - Apical periodontitis represents a chronic inflammatory process within periapical tissues, mostly caused by etiological agents of endodontic origin. Progressive bone resorption in the periapical region represents the hallmark of apical periodontitis and occurs as the consequence of interplay between polymicrobial infections and host response. The Notch signaling pathway is an evolutionary conserved cell-signaling system that plays an important role in a variety of cell functions including proliferation, differentiation and apoptosis. In recent years its involvement in bone homeostasis has attracted a significant consideration. We hypothesized that Notch signaling pathway, which has a complex interplay with proinflammatory cytokines and bone resorption regulators, contributes to alveolar bone resorption via increased Notch receptors on immune cell surface and stimulates Notch receptor intracellular domain (NICD) translocation into the nucleus. The potential benefit of medications aimed to down-regulate these pathways in apical periodontitis treatment remains to be assessed.

This study aimed to analyze the expression of Notch signaling molecules (Notch2, Jagged1, and Hey1) and proinflammatory cytokines (tumor necrosis factor alpha [TNF-alpha], interleukin [IL]-1 beta, and IL-6) in human apical periodontitis lesions with different receptor activator of nuclear factor kappa B ligand (RANKL)/osteo-protegerin (OPG) ratios and determine their potential correlation. Methods: The study group consisted of 50 periapical lesions collected in conjunction with apicoectomy. The relative gene expression of the investigated molecules (Notch2, Jagged1, Hey1, RANKL, OPG, TNF-alpha, IL-1 beta, and IL-6) in all tissue samples was analyzed using reverse transcriptase real-time polymerase chain reaction. The Student t test, Mann-Whitney U test and Spearman correlation were used for statistical analysis. Results: Based on the RANKUOPG ratio, periapical lesions were either RANKL predominant (RANKL > OPG, n = 33) or OPG predominant (RANKL  lt  OPG, n = 17). Symptomatic lesions occurred more frequently in RANKL-predominant compared with OPG-predominant lesions (24 vs 7, P=.029). Notch2,Jagged1, Hey1, and TNF-alpha were significantly overexpressed in lesions with predominant RANKL compared with lesions with predominant OPG (P =.001, P =.001, P =.027, and P =.016, respectively). Significant correlations were observed between the investigated genes in periapical lesions. Conclusions: Notch signaling appeared to be activated in periapical inflammation. An increase in Notch2, Jagged1, Hey1, and TNF-alpha expression in RANKL-predominant periapical lesions corroborates their joined involvement in extensive periapical bone resorption.

Contact angles of reference liquids and SFE of dentin following He + 1.5% O-2 at 3-W and 4-mm treatment was compared to phosphoric acid etching. Contact angles of Single Bond Universal (SBU; 3M ESPE) and Clearfil Universal Bond (CUB; Kuraray Noritake) were measured following NTAP, etch-and-rinse, and self-etch protocols.ResultsNTAP significantly reduced contact angles of reference liquids and increased dentin SFE compared to untreated dentin (p lt 0.05). O-2 intensified the effect of He NTAP (p lt 0.05). NTAP and phosphoric acid increased dentin polarity and Lewis base surface characteristics. Phosphoric acid increased contact angles of adhesives compared to the self-etch protocol (p lt 0.05). AB  - ObjectivesThe study aims to evaluate the effects of non-thermal atmospheric plasma (NTAP) treatments on dentin wetting and surface free energy (SFE) and compare the effects of NTAP treatment, etch-and-rinse, and self-etch protocols for application of universal adhesives.Materials and methodsMid-coronal dentin of intact third molars was used to measure contact angles of distilled water, ethylene-glycol, and diiodomethane and calculate SFE following different NTAP preset treatments (feeding gas consisting of pure He, He + 1% O-2, He + 1.5% O-2), power input (1 or 3W), and tip-to-surface distance (2, 4, or 8mm). Contact angles of reference liquids and SFE of dentin following He + 1.5% O-2 at 3-W and 4-mm treatment was compared to phosphoric acid etching. Contact angles of Single Bond Universal (SBU; 3M ESPE) and Clearfil Universal Bond (CUB; Kuraray Noritake) were measured following NTAP, etch-and-rinse, and self-etch protocols.ResultsNTAP significantly reduced contact angles of reference liquids and increased dentin SFE compared to untreated dentin (p lt 0.05). O-2 intensified the effect of He NTAP (p lt 0.05). NTAP and phosphoric acid increased dentin polarity and Lewis base surface characteristics. Phosphoric acid increased contact angles of adhesives compared to the self-etch protocol (p lt 0.05). NTAP resulted in lower adhesive contact angles than phosphoric acid, the difference being statistically significant for CUB (p lt 0.05). Compared to the self-etch protocol, NTAP slightly reduced CUB contact angle but not that of SBU (p>0.05).ConclusionsHe NTAP with and without O-2 increased dentin wetting and SFE, surpassing the effect of phosphoric acid and lowering adhesive contact angles. This review describes the most recent findings on herpesviral infections and offers current concepts of herpesviral role in the pathogenesis of periapical periodontitis.

Recent Findings
Thirty articles reported data on herpesviral infection in periapical periodontitis. Epstein-Barr virus and human cytomegalovirus are the most frequently detected herpesviruses in periapical samples. The main hypothesis postulates a bidirectional herpesviral-bacterial relationship in the etiopathogenesis of periapical periodontitis. A high heterogeneity of herpesviruses incidence was registered within the studies, in part, due to various methodological approaches used in laboratory testing, different inclusion criteria, study design, seroprevalence of herpesviruses, and sociodemographic characteristics of investigated populations.

Summary
Herpesviruses have been shown to potentially impair local host defense in periapical tissue. Although it has been demonstrated that endodontic pathogenic bacteria are able to reactivate herpesviruses, further, in vitro studies should provide more data on herpesviruses as a factor in the pathogenesis of the periapical pathoses. It is, therefore, necessary to investigate potential benefits of antiviral therapy in well-designed controlled longitudinal studies.

MethodologyThe experimental group consisted of 30 EBV-positive and 30 EBV-negative periapical lesions collected in conjunction with apicoectomy. The pulp tissues of 20 impacted third molars were used as healthy controls. The qualitative and quantitative analysis of EBV was performed by nested and real-time polymerase chain reaction (PCR), respectively. The levels of RANKL and OPG were analysed by reverse transcriptase real-time PCR. The levels of 8-OHdG and GSSG were determined by enzyme-linked immunosorbent assay (ELISA). Mann-Whitney U-test and Spearman's correlation were used for statistical analysis. ResultsThe levels of RANKL, OPG, 8-OHdG and GSSG were significantly higher in apical periodontitis lesions compared to healthy pulp controls (P=0.001, P lt 0.001, P lt 0.001 and P lt 0.05, respectively). RANKL and OPG mRNA expression was significantly higher in EBV-positive compared to EBV-negative periapical lesions (P lt 0.05). There was no significant correlation between EBV copy numbers and levels of RANKL, OPG, 8OH-dG and GSSG in apical periodontitis. ConclusionLevels of bone resorption regulators and oxidative stress biomarkers were increased in apical periodontitis compared to healthy pulp tissues. AB  - The pathogenesis of obesity-related vascular disorders has not been fully elucidated. The fundamental role of inflammation in aging process is now widely recognized, particularly for atherosclerotic disease which begins before birth. The number of obese individuals worldwide has reached two billion, leading to an explosion of obesity-related vascular disorders associated with increased morbidity and mortality. Obesity, as a chronic low grade inflammatory process, is important risk factor for metabolic and cardiovascular disease. Despite a well-known genetic component, this risk appears to originate from several abnormalities in adipose tissue function associated with a chronic inflammatory state. In particular, obesity as the most common nutritional disorder in industrialized countries, is closely related to impaired endothelial function, a well-known marker of preatherosclerotic disease. These conditions disrupt vascular homeostasis by causing an imbalance between the nitric oxide pathway and the endothelin-1 system, with impaired insulin- stimulated endothelium-dependent vasodilation. Having in mind the growing population of overweight and obese people worldwide, along with an increasingly aging population, understanding the pathophysiology of obesity on cardiovascular system is essential. The mechanisms linking obesity-related vascular disorders and low grade inflammation in aging process are the focus of this paper.
AB  - Patogeneza vaskularnih poremećaja koji su povezani sa gojaznošću nije u potpunosti razjašnjena. Danas je poznata fundamentalna uloga inflamacije u procesu starenja, naročito kod aterosklerozne bolesti, koja počinje pre rođenja. Broj gojaznih osoba širom sveta je dostigao cifru dva biliona i doveo do “eksplozije” vaskularnih poremećaja koji su povezani sa gojaznošću i povećanim morbiditetom i mortalitetom. Gojaznost, kao hronični niskostepeni inflamacijski proces, je važan faktor rizika za nastanak metaboličkih i kardiovaskularnih bolesti. Pored dobro proučene genetske komponente, izgleda da je za ovaj rizik zaslužno i nekoliko poremećaja funkcije masnog tkiva koji se dovode u vezu sa stanjem hronične inflamacije. U tom smislu prednjači gojaznost, pošto je kao najčešći poremećaj ishrane u razvijenim zemljama blisko povezana sa endotelnom disfunkcijom, koja predstavlja uveliko poznat marker preaterosklerozne bolesti. Takvi uslovi narušavaju vaskularnu homeostazu, dovodeći do dizbalansa između metaboličkog puta azot monoksida i sistema endotelina-1, što je praćeno otežanim odvijanjem insulinom podstaknute vazodilatacije. Imajući u vidu globalni zajednički porast broja kako osoba sa viškom kilograma i gojaznih osoba, tako i starije populacije, razumevanje patogeneze gojaznosti i njenog uticaja na kardiovaskularni sistem je od esencijalnog značaja. Mehanizmi koji čine sponu između vaskularnih poremećaja povezanih sa gojaznošću i niskostepene inflamacije u procesu starenja nalaze se u fokusu ovog rada.

These conditions disrupt vascular homeostasis by causing an imbalance between the nitric oxide pathway and the endothelin-1 system, with impaired insulin- stimulated endothelium-dependent vasodilation. Having in mind the growing population of overweight and obese people worldwide, along with an increasingly aging population, understanding the pathophysiology of obesity on cardiovascular system is essential. The mechanisms linking obesity-related vascular disorders and low grade inflammation in aging process are the focus of this paper., Patogeneza vaskularnih poremećaja koji su povezani sa gojaznošću nije u potpunosti razjašnjena. Danas je poznata fundamentalna uloga inflamacije u procesu starenja, naročito kod aterosklerozne bolesti, koja počinje pre rođenja. Broj gojaznih osoba širom sveta je dostigao cifru dva biliona i doveo do “eksplozije” vaskularnih poremećaja koji su povezani sa gojaznošću i povećanim morbiditetom i mortalitetom. Gojaznost, kao hronični niskostepeni inflamacijski proces, je važan faktor rizika za nastanak metaboličkih i kardiovaskularnih bolesti. Pored dobro proučene genetske komponente, izgleda da je za ovaj rizik zaslužno i nekoliko poremećaja funkcije masnog tkiva koji se dovode u vezu sa stanjem hronične inflamacije. U tom smislu prednjači gojaznost, pošto je kao najčešći poremećaj ishrane u razvijenim zemljama blisko povezana sa endotelnom disfunkcijom, koja predstavlja uveliko poznat marker preaterosklerozne bolesti. Takvi uslovi narušavaju vaskularnu homeostazu, dovodeći do dizbalansa između metaboličkog puta azot monoksida i sistema endotelina-1, što je praćeno otežanim odvijanjem insulinom podstaknute vazodilatacije. Imajući u vidu globalni zajednički porast broja kako osoba sa viškom kilograma i gojaznih osoba, tako i starije populacije, razumevanje patogeneze gojaznosti i njenog uticaja na kardiovaskularni sistem je od esencijalnog značaja. Mehanizmi koji čine sponu između vaskularnih poremećaja povezanih sa gojaznošću i niskostepene inflamacije u procesu starenja nalaze se u fokusu ovog rada.",

Methods: The literature search covered MEDLINE, Science Citation Index Expanded, and Scopus. A qualitative critical appraisal of the included articles was performed. Results: The electronic database search. yielded 66 hits from PubMed, 73 hits from Web of Science, and 107 from Scopus: Seven case reports and 3 cross-sectional studies were included in the final review. When summarized, in 7 patients with a history of a previous HZ attack and with no other apparent cause, -23 teeth were diagnosed with apical periodontitis, 8 teeth with internal and 1 tooth with external root resorption. The cross-sectional studies investigated the presence of VZV DNA in samples of acute apical abscess. The VZV DNA was found only in 2 of 65 samples. Conclusions: All studies included in this systematic review.had a low level of evidence (4 and 5). Still, the potential role of VZV in the etiopathogenesis of periapical pathoses and root resorption cannot be ruled out. Future investigations should be directed toward the analysis of VZV pathologic effects on pulp blood vessels, which might cause local ischemia and tissue necrosis.

AB  - Apical periodontitis represents a chronic inflammation and destruction of periradicular tissue caused by polymicrobial infection of endodontic origin. The aim of this systematic review was to make an update on findings related to Epstein-Barr virus (EBV) and human cytomegalovirus (HCMV) presence in periapical pathoses and to correlate these findings with clinical, histopathological and radiographic features of periapical lesions. Methods were based on the preferred reporting items for systematic reviews and meta-analyses (PRISMA) statement. A search was performed using PubMed, Web of Science and SCOPUS. Search key words included the following medical subjects heading terms: (periapical disease OR apical periodontitis OR periapical lesions OR periapical abscess) AND (viruses OR herpesvir*). A manual search involved references from articles retrieved for possible inclusion. The search, evalua‐ tion, and critical appraisal of articles were performed by two independent judges. Collected data were analyzed using the measures of descriptive statistics. The final review has included twenty nine articles related to herpesviral presence periapical pathoses. Qualitative analysis indicated that EBV HCMV, and HHV-8 were the most prevalent species in periapical pathoses. Our findings suggest that there is wide variety of herpesviruses detection rates in periapical pathoses in relation to their clinical, histopathological and radiographic features.

Apart from bacteria, two herpesviruses, Epstein-Barr virus (EBV) and Human cytomegalovirus (HCMV) are recognized as putative pathogens in apical periodontitis. Although previous reports suggest the involvement of EBV in the pathogenesis of apical periodontitis, its exact role in periapical bone resorption has not yet been fully elucidated. We hypothesize that EBV infection in apical periodontitis is capable of inducing periapical bone resorption via stimulation of reactive oxygen species (ROS) overproduction. Increased levels of ROS induce expression of receptor activator of nuclear factor kappa B (NF-kappa B) ligand (RANKL). RANKL binding to receptor activator of nuclear factor kappa B (RANK) present on the surface of preosteoclasts induces their maturation and activation which consequently leads to bone resorption. The potential benefit of antiviral and antioxidant-based therapies in periapical bone resorption treatment remains to be assessed.

AB  - In this paper we will present results for plasma sterilization of planktonic samples of two reference strains of bacteria, Pseudomonas aeruginosa ATCC 27853 and Enterococcus faecalis ATCC 29212. We have used a plasma needle as a source of non-equilibrium atmospheric plasma in all treatments. This device is already well characterized by OES, derivative probes and mass spectrometry. It was shown that power delivered to the plasma is bellow 2 W and that it produces the main radical oxygen and nitrogen species believed to be responsible for the sterilization process. Here we will only present results obtained by electron paramagnetic resonance which was used to detect the OH, H and NO species. Treatment time and power delivered to the plasma were found to have the strongest influence on sterilization. In all cases we have observed a reduction of several orders of magnitude in the concentration of bacteria and for the longest treatment time complete eradication. A more efficient sterilization was achieved in the case of gram negative bacteria.

AB  - Mesenchymal stem cells (MSCs) were isolated from human periodontal ligament (hPDL-MSCs) and characterized by their morphology, clonogenic efficiency, proliferation and differentiation capabilities. hPDL-MSCs, derived from normal impacted third molars, possessed all of the properties of MSC, including clonogenic ability, high proliferation rate and multi-lineage (osteogenic, chondrogenic, adipogenic, myogenic) differentiation potential. Moreover, hPDL-MSCs expressed a typical MSC epitope profile, being positive for mesenchymal cell markers (CD44H, CD90, CD105, CD73, CD29, Stro-1, fibronectin, vimentin, alpha-SMA), and negative for hematopoietic stem cell markers (CD34, CD11b, CD45, Glycophorin-CD235a). Additionally, hPDL-MSCs, as primitive and highly multipotent cells, showed high expression of embryonic markers (Nanog, Sox2, SSEA4). The data obtained provided yet further proof that cells with mesenchymal properties can be obtained from periodontal ligament tissue. Although these cells should be further investigated to determine their clinical significance, hPDL-MSCs are believed to provide a renewable and promising cell source for new therapeutic strategies in the treatment of periodontal defects.

In this paper we present a study of the effectiveness of an atmospheric pressure plasma source, known as plasma needle, in inhibition of the growth of biofilm produced by methicillin resistant Staphylococcus aureus (MRSA). Even at the lowest powers the biofilms formed by inoculi of MRSA of 10(4) and 10(5) CFU have been strongly affected by plasma and growth in biofilms was inhibited. The eradication of the already formed biofilm was not achieved and it is required to go to more effective sources.

AB  - Here we investigate the influences of non-thermal atmospheric plasma on human mesenchymal stem cells isolated from periodontal ligament (hPDL-MSCs). A specially redesigned plasma needle was used as the source of low-temperature plasma and its effects on different hPDL-MSC functions were investigated. Cell cultures were obtained from extracted normal impacted third molars and characterized for their phenotype and multi-potential differentiation. The hPDL-MSCs possessed all the typical MSC properties, including clonogenic ability, high proliferation rate, specific phenotype and multilineage differentiation. The data regarding the interaction of plasma with hPDL-MSCs demonstrated that plasma treatment inhibited the migration of hPDL-MSCs and induced some detachment, while not affecting their viability. Additionally, plasma significantly attenuated hPDL-MSCs' proliferation, but promoted their osteogenic differentiation. The results of this study indicated that a non-thermal plasma offers specific activity with non-destructive properties that can be advantageous for future dental applications.

AB  - Background & objectives: The interleukin (IL)-17 producing T-helper cells have been linked to pathogenesis of autoimmunity and mostly investigated in rheumatoid arthritis (RA). In this study we tested the IL-17 levels, as well as the levels of nitric oxide (NO) as possible IL-17-induced product, in patients with primary Sjogren's syndrome (pSS), an intricate and complex chronic autoimmune disorder of exocrine glands. Methods: Serum IL-17 levels and nitrite concentrations determined in patients with pSS (n=30) were compared with the values obtained in patients with RA (n=10) and healthy controls (n=15). The values obtained for 11,17 in pSS patients were also associated with the patients' clinical characteristics, particularly the rheumatoid factor (RF) and total antinuclear antibodies (tANA) levels. Results: Serum concentrations of IL-17 were significantly (P lt 0.01) higher in patients with pSS (12.9 +/- 28.0 pg/ml) as compared to those obtained in healthy individuals (0.2 +/- 0.6 pg/ml), but not as high as the values obtained for the patients with RA (34.5 +/- 56.2 pg/ml). The mean IL-17 levels were significantly (P lt 0.05) higher in the pSS patients positive for rheumatoid factor (20.3 +/- 33.3 pg/ml) than in RF-negatives (0.3 +/- 0.6 pg/ml). Mean serum concentrations of IL-17 were also higher in antinuclear antibody (ANA)positive samples (19.8 +/- 33.5 pg/ml) in comparison to ANA-negative sera (1.1 +/- 3.1 pg/ml) (P lt 0.05). The NO levels also showed elevated values in both pSS and RA patients, as compared to the healthy controls, since mean nitrite levels in patients with pSS and RA were 38.2 +/- 29.2 mu M and 41.7 +/- 21.1 mu M, respectively, while those in healthy controls were significantly lower, at 19.2 +/- 10.5 mu M. Interpretation & conclusions: The findings of this study showed that there was increased IL-17 and NO production in patients with primary SS, especially if they had associated elevated rheumatoid factor and antinuclear antibody values.

AB  - The potential of plasma applications medicine, the connections to nanotechnologies and the results obtained by our group are reviewed. A special issue in plasma medicine is the development of the plasma sources that would achieve non-equilibrium at atmospheric pressure in an atmospheric gas mixture with no or only marginal heating of the gas, and with desired properties and mechanisms that may be controlled. Our studies have shown that control of radicals or chemically active products of the discharge, such as ROS (reactive oxygen species) and/or NO, may be used to control the growth of the seeds. Simultaneously, a specially designed plasma needle and other sources were shown to be efficient to sterilize not only colonies of bacteria but also plank- tonic samples (microorganisms protected by water) or bio films. Finally, it was shown that a plasma might induce differentiation of stem cells. Non-equilibrium plasmas may be used in detection of different specific markers in medicine. For example proton transfer mass spectroscopy may be employed in the detection of volatile organic compounds without their dissociation and thus as a technique for instantaneous measurement of the presence of markers for numerous diseases.
AB  - U ovom radu dat je pregled primene plazme u medicini, povezanost sa nanotehnologijama i rezultate na ovom polju koje je postigla naša grupa. Poseban problem u plazma medicini je razvoj izvora plazme koji bi radili u neravnotežnim uslovima na atmosferskom pritisku i u smeši gasova kakva je u atmosferi uz zanemarljivo grejanje gasa i sa željenim karakteristikama koje se mogu podešavati po želji. Naša istraživanja su pokazala da se kontrola prisustva radikala i drugih hemijski aktivnih čestica kao što su reaktivne kiseonične čestice (ROS) i/ili NO, može koristiti za kontrolu klijanja semenki. U isto vreme je dokazano za posebno konstruisanu plazma iglu da može efikasno da steriliše ne samo kolonije bakterija već i planktonske uzorke (mikroorganizme zaštićene vodom) pa i biofilmove. Na kraju, mi smo pokazali da plazma može da indukuje diferencijaciju matičnih ćelija. Neravnotežna plazma se može koristiti za detekciju raznih specifičnih markera u medicini. Na primer masena spektroskopija na bazi izmene protona može da se koristi za detekciju isparivih organskih jedinjenja bez njihove disocijacije i na taj način se može ostvariti trenutna detekcija markera za brojne bolesti iz daha.

Na kraju, mi smo pokazali da plazma može da indukuje diferencijaciju matičnih ćelija. Neravnotežna plazma se može koristiti za detekciju raznih specifičnih markera u medicini. Na primer masena spektroskopija na bazi izmene protona može da se koristi za detekciju isparivih organskih jedinjenja bez njihove disocijacije i na taj način se može ostvariti trenutna detekcija markera za brojne bolesti iz daha.",

AB  - In this paper, we study the application of a plasma needle to induce necrosis in planktonic samples containing a single breed of bacteria. Two different types of bacteria, Staphylococcus aureus (ATCC 25923) and Escherichia coli (ATCC 25922), were covered in this study. In all experiments with bacteria, the samples were liquid suspensions of several different concentrations of bacteria prepared according to the McFarland standard. The second system studied in this paper was human peripheral blood mesenchymal stem cells (hPB-MSC). In the case of hPB-MSC, two sets of experiments were performed: when cells were covered with a certain amount of liquid (indirect) and when the cell sample was in direct contact with the plasma. Most importantly, the study is made with the aim to see the effects when the living cells are in a liquid medium, which normally acts as protection against the many agents that may be released by plasmas. It was found that a good effect may be expected for a wide range of initial cell densities and operating conditions causing destruction of several orders of magnitude even under the protection of a liquid. It was established independently that a temperature increase could not affect the cells under the conditions of our experiment, so the effect could those hPB-MSC that were not protected by a liquid, gas flow proved to produce a considerable effect, presumably due to poor adhesion of the cells, but in a liquid the effect was only due to the plasma. Further optimization of the operation may be attempted, opening up the possibility of localized in vivo sterilization.
