AB  - Objective: We aimed to investigate alteration in cellular signaling mediated by vascular endothelial growth factor (VEGF) and parameters of oxidative stress/nitric oxide generation, superoxide dismutase (SOD) and neuronal nitric oxide synthase (nNOS), underlying altered functional mechanical loading of TMJ (temporomandibular joint) during lateral mandibular deviation. Design: Thirty-eight 5-week-old male Wistar rats were divided into experimental group, which received acrylic resin appliance that shifted mandible to the left during closure, and control group. Computed tomography and histomorphometry were used for condyle analyses, while samples of condyle, synovial membrane and m. masseter were analyzed with enzyme-linked immunosorbent assay and spectrophotometry to determine VEGF and nNOS protein concentrations, and SOD activity. Results: Experimental group of rats developed smaller and asymmetrical mandibles. Less of new bone and cartilage formation and larger bone marrow cavities area were found in the experimental group. Higher VEGF expression in condyle and m. masseter as well as higher nNOS expression in m. masseter and synovial membrane were found in the experimental compared to the control group. Alteration of SOD activity was found in m. masseter and synovial membrane in the experimental group. Conclusions: Lateral mandibular deviation induces mandibular and condylar morphological changes as well as significant cellular signaling alterations in condyle, synovial membrane and masticatory muscle. Cellular VEGF protein overexpression and oxidative stress/nitric oxide disbalance could be the mechanisms underlying unbalanced functional TMJ loading due to mandibular deviation.

Subchronic Systemic Toxicity of New Endodontic Material Based on Calcium Hydroxyapatite and Calcium Silicates

AB  - As an alternative to MTA, a new endodontic material based on hydroxyapatite and calcium silicates (ALBO-MPSA) has been synthesized, and its biocompatibility has been studied in many in vitro and in vivo studies. The current study aims to evaluate a subchronic systemic toxicity of ALBO-MPSA on the rat animal model, as a continuation of the previous studies. Biochemical parameters of blood and histological parameters of the liver, kidneys, and spleen of the rats were analyzed after 120 days of consumption of the aqueous extract of ALBO-MPSA. The results showed no myelotoxic effect or autoimmune effect on peripheral blood cells and no pathological effect on the liver, kidney, and spleen tissues. Besides, no changes in the skin and hair of the rats, neither the change in the consumption of food and water, nor the change in their usual behavior were noticed during the experiment.

Nanostructured endodontic materials mixed with different radiocontrast agentsbiocompatibility study

AB  - The aim of this study was to investigate the biocompatibility of nanostructured materials based on highly active calcium silicates mixed with different radiocontrast agents in comparison to MTA(+) using in vitro and in vivo model. Morphology of materials' samples was analyzed using SEM while the phase compositions were identified by XRD. pH values of materials' suspensions were conducted by pH-meter. The cytotoxicity of materials' solutions was tested by MTT test (100, 50, 25 and 12.5mg/ml). LDH and H-3-thymidine assay were utilized for biocompatibility investigations of materials' eluates (24h, 7 day and 21 day). Eighteen Guinea pigs were used for intramuscular implantation, as teflon tubes with freshly prepared materials were placed into intramuscular pockets. All samples were composed of round and needle-like particles equally distributed with Ca/Si ratio 2.7 at%, with the presence of hydrated calcium silicate phases. The pH values of ALBO-MPCA(1) and ALBO-MPCA(2) were high alkaline, while in case of MTA(+) they were lower and continuously declined (p lt 0.05). Investigated materials didn't exhibit dose-dependent effect on metabolic activity of L929 cells (p>0.05). Significant differences in the percentage of cytotoxicity between diluted and undiluted extracts between all tested materials after 24h and 7 day were noticed (p lt 0.05). Increase in L929 cells proliferation was noticed in case of undiluted eluates of ALBO-MPCA(1) and ALBO-MPCA(2) after 7 day (p lt 0.05). There were no statistically significant differences in the intensity of inflammatory response between investigated materials and control group after 60 day (p>0.05). Evaluation of biocompatibility of both ALBO-MPCA(1) and ALBO-MPCA(2) indicate their potential clinical use. [GRAPHICS] .

AB  - Introduction: The aim of this article was to analyze biocompatibility and bioactivity of new endodontic materials on the basis of nanosynthesized calcium silicates (ALBO-MPCA(1) and ALBO-MPCA(2)) combined with different radiopacifiers in comparison with MTA(+). Methods: Morphology of the samples was studied by scanning electron microscopy, and the pH and ion release analysis were also assessed. Biocompatibility of materials' eluates (24-hour, 7-day, and 21-day) was conducted by using MIT test. Twelve New Zealand white rabbits were used for intraosseous implantation. Four calvarial defects per animal were created and filled with freshly prepared investigated materials. Results: Samples mostly consisted of agglomerates built up from nanoparticles, preferably spherical and rod-like. There was no significant difference among pH values of materials' eluates after 24 hours (P>.05). The amount of calcium and aluminum ion release decreased, whereas the amount of magnesium and bismuth (ALBO-MPCAl, MTA(+)) and barium (ALBO-MPCA(2)) increased during 21-day period. The metabolic activity of cells increased after the extraction time, except in case of undiluted elutes of ALBO-MPCA(2) and ALBO-MPCAI (21-day). Histologic analysis of the samples revealed newly formed bone tissue with moderate inflammation for all investigated materials, which subsided during 90-day period to mild. Both MIA(+) and ALBO-MPCAI were in direct contact with the newly formed bone tissue. After 90 days, statistically significant difference in hard tissue formation was observed in comparison of MIA(+) and ALBO-MPCki with control group (P  lt .05). Conclusions: Experimental materials ALBOMPCA, and ALBO-MPCA(2) possess both biocompatibility and bioactivity. Because ALBO-MPCAA provokes favorable biological response, it is especially good candidate for further clinical investigations.

AB  - Background/Aim. The mechanism of impaired bone healing in diabetes mellitus includes different tissue and cellular level activities due to micro- and macrovascular changes. As a chronic metabolic disease with vascular complications, diabetes affects a process of bone regeneration as well. The therapeutic approach in bone regeneration is based on the use of osteoinductive autogenous grafts as well as osteoconductive synthetic material, like a β-tricalcium phosphate. The aim of the study was to determine the quality and quantity of new bone formation after the use of autogenous bone and β-tricalcium phosphate in the model of calvarial critical-sized defect in rabbits with induced diabetes mellitus type I. Methods. The study included eight 4-month-old Chincilla rabbits with alloxan-induced diabetes mellitus type I. In all animals, there were surgically created two calvarial bilateral defects (diameter 12 mm), which were grafted with autogenous bone and β-tricalcium phosphate (n = 4) or served as unfilled controls (n = 4). After 4 weeks of healing, animals were sacrificed and calvarial bone blocks were taken for histologic and histomorphometric analysis. Beside descriptive histologic evaluation, the percentage of new bone formation, connective tissue and residual graft were calculated. All parameters were statistically evaluated by Friedman Test and post hock Wilcoxon Singed Ranks Test with a significance of p  lt  0.05. Results. Histology revealed active new bone formation peripherally with centrally located connective tissue, newly formed woven bone and well incorporated residual grafts in all treated defects. Control samples showed no bone bridging of defects. There was a significantly more new bone in autogeonous graft (53%) compared with β-tricalcium phosphate (30%), (p  lt  0.030) and control (7%), (p  lt  0.000) groups. A significant difference was also recorded between β-tricalcium phosphate and control groups (p  lt  0.008). Conclusion. In the present study on the rabbit grafting model with induced diabetes mellitus type I, the effective bone regeneration of critical bone defects was obtained using autogenous bone graft.
AB  - Uvod/Cilj. Mehanizam otežanog zarastanja tkiva kod dijabetesa melitusa zasnovan je na različitim promenama funkcije na tkivnom i ćelijskom nivou, usled prisutnih mikro- i makrovaskularnih promena. Kao hronično metaboličko oboljenje sa vaskularnim komplikacijama, dijabetes melitus zahvata i proces koštane regeneracije. Terapijski postupci u okviru regeneracije kosti obuhvataju primenu autotransplantata sa oseoinduktivnim delovanjem i sintetskih osteokonduktivnih materijala, kao što je i β-trikalcijum fosfat. Cilj ovog rada bio je da se ispita kvantitet i kvalitet novoformiranog koštanog tkiva posle korišćenja autotransplantata kosti i β-trikalcijum fosfata, na modelu kritičnog defekta kalvarije kunića sa eksperimentalno izazvanim dijabetesom melitusom tipa I. Značajno više novoformirane kosti bilo je prisutno u defektima regenerisanim autotransplantatom (53%) u poređenju sa kontrolnim defektima (7%), (p  lt  0,000) i defektima popunjenim β-trikalcijum fosfatom (30%), (p  lt  0,030). Takođe, značajna razlika uočena je i između grupe sa β-trikalcijum fosfatom i kontrolnim koštanim defektom (p  lt  0,008). Zaključak. Primena autotransplantata kosti značajno povećava uspešnost regeneracije kritičnih defekata kosti kalvarije kunića sa dijabetesom melitusom tipa I.

Histological effects of enamel matrix derivative on exposed dental pulp

AB  - Introduction Direct pulp capping procedure is a therapeutic application of a drug on exposed tooth pulp in order to ensure the closure of the pulp chamber and to allow the healing process to take place. Objective The aim of this study was to examine the histological effects of Emdogain® on exposed tooth pulp of a Vietnamese pig (Sus scrofa verus). Methods The study comprised 20 teeth of a Vietnamese pig. After class V preparation on the buccal surfaces of incisors, canines and first premolars, pulp was exposed. In the experimental group, the perforations were capped with Emdogain® (Straumann, Basel, Switzerland), while in the control group pulp capping was performed with MTA® (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass-ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). The observational period was 28 days, after which the animal was sacrificed and histological preparations were made. A light microscope was used to analyze dentin bridge formation, tissue reorganization and inflammation, and the presence of bacteria in the pulp. Results The formation of dentin bridge was observed in the experimental and control groups. Inflammation of the pulp was mild to moderate in both groups. Angiogenesis and many odontoblast-like cells, responsible for dentin bridge formation, were observed. Necrosis was not observed in any case, nor were bacteria present in the pulp. Conclusion Histological analysis indicated a favorable therapeutic effect of Emdogain® Gel in direct pulp capping of Vietnamese pigs. Pulp reaction was similar to that of MTA®.
AB  - Uvod Direktno prekrivanje pulpe je važan terapijski postupak primene leka na eksponiranu pulpu zuba, čiji je cilj da obezbedi zatvaranje pulpne komore i omogući proces zarastanja. Cilj rada Cilj ovog rada je bio da se histološki ispita efekat gleđnog emdogaina na eksponiranu pulpu zuba vijetnamske svinje (Sus scrofa domesticus). Metode rada Istraživanje je urađeno na 20 zuba vijetnamske svinje. Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase s eksponiranjem komora pulpe. U eksperimentalnoj grupi perforacija je prekrivana proizvodom Emdogain® (Straumann, Bazel, Švajcarska), a u kontrolnoj sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer-cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Period posmatranja trajao je 28 dana, a nakon žrtvovanja životinja, napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, zapaljenjska reakcija pulpe, reorganizacija pulpnog tkiva i postojanje bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe uočeno je stvaranje dentinskog mostića. Upala pulpe je bila blaga do umerena i u eksperimentalnoj i u kontrolnoj grupi. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo bakterija u pulpi. Direct pulp capping using Biodentine

AB  - Introduction Direct pulp capping is therapeutic method of applying medication on exposed pulp in order to allow bridge formation and healing process. The aim of this study was to investigate the effect of Biodentine on exposed dental pulp of Vietnamese pigs. Material and Methods The study was conducted on 20 teeth of Vietnamese pigs (Sus scrofa domesticus). On buccal surfaces of incisors, canines and first premolars, class V cavities were prepared and pulp was exposed. In the experimental group (six incisors, two canines and two premolars) the perforation was covered with BiodentineR (Septodont, Saint-Maur-des-Fosses, France). In the control group, the perforation was covered with MTAR (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, an inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in both groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of Biodentine for direct pulp capping in teeth of Vietnamese pigs. Findings were similar with Biodentine and MTA.
AB  - Uvod Direktno prekrivanje pulpe je terapijski postupak primene leka na eksponiranu pulpu zuba radi zatvaranja pulpne komore i omogućavanja procesa zarastanja. Cilj ovog rada je bio da se ispita efekat biodentina na eksponiranu pulpu zuba vijetnamske svinje. Materijal i metode rada Istraživanje je izvršeno na 20 zuba vijetnamske svinje (Sus scrofa domesticus). Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase, pri čemu je napravljena namerna eksploracija komora pulpe. U eksperimentalnoj grupi (šest sekutića, dva očnjaka i dva premolara) perforacija je prekrivana preparatom Biodentine ® (Septodont, Sent Mor de Fos, Francuska). U kontrolnoj grupi perforacija je prekrivana sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer- cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Opservacioni period trajao je 28 dana. Nakon žrtvovanja životinja napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, inflamatorna reakcija pulpe, reorganizacija pulpnog tkiva i prisustvo bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe zabeleženo je stvaranje dentinskog mostića. Zapaljenje pulpe je bilo blago do umereno i u eksperimentalnoj i u kontrolnoj grupi uzoraka. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo Gram-pozitivnih bakterija u pulpi. abstract = "Introduction Direct pulp capping is therapeutic method of applying medication on exposed pulp in order to allow bridge formation and healing process. The aim of this study was to investigate the effect of Biodentine on exposed dental pulp of Vietnamese pigs. Material and Methods The study was conducted on 20 teeth of Vietnamese pigs (Sus scrofa domesticus). On buccal surfaces of incisors, canines and first premolars, class V cavities were prepared and pulp was exposed. In the experimental group (six incisors, two canines and two premolars) the perforation was covered with BiodentineR (Septodont, Saint-Maur-des-Fosses, France). In the control group, the perforation was covered with MTAR (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, an inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in both groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of Biodentine for direct pulp capping in teeth of Vietnamese pigs. Findings were similar with Biodentine and MTA., Uvod Direktno prekrivanje pulpe je terapijski postupak primene leka na eksponiranu pulpu zuba radi zatvaranja pulpne komore i omogućavanja procesa zarastanja. Cilj ovog rada je bio da se ispita efekat biodentina na eksponiranu pulpu zuba vijetnamske svinje. Materijal i metode rada Istraživanje je izvršeno na 20 zuba vijetnamske svinje (Sus scrofa domesticus). Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase, pri čemu je napravljena namerna eksploracija komora pulpe. U eksperimentalnoj grupi (šest sekutića, dva očnjaka i dva premolara) perforacija je prekrivana preparatom Biodentine ® (Septodont, Sent Mor de Fos, Francuska). U kontrolnoj grupi perforacija je prekrivana sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer- cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Opservacioni period trajao je 28 dana. Nakon žrtvovanja životinja napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, inflamatorna reakcija pulpe, reorganizacija pulpnog tkiva i prisustvo bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe zabeleženo je stvaranje dentinskog mostića. Zapaljenje pulpe je bilo blago do umereno i u eksperimentalnoj i u kontrolnoj grupi uzoraka. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo Gram-pozitivnih bakterija u pulpi. Zaključak Histološka analiza je ukazala na povoljne terapijske efekte biodentina u direktnom prekrivanju pulpe zuba vijetnamskih svinja. Reakcija pulpe bila je slična onima koje je izazvao MTA.",

Histological evaluation of direct pulp capping with novel nanostructural materials based on active silicate cements and biodentine® on pulp tissue

In the control group, exposed pulp was covered with ProRoot MTA® (10 teeth). After the observation period of 28 days, the animals were sacrificed and the teeth prepared for histological analysis. Light microscope was used for the analysis of dentin bridge formation, tissue reorganization and inflammation, and the presence of bacteria in the pulp. In the group of Biodentine®, a complete dentin bridge was noted in 3 cases, while incomplete dentin bridge in the form of dental islets was detected in 4 cases. Nanostructured material ALBO-MPCA I provided complete dentin bridge formation in 5 teeth, in 3 teeth the formed dentin bridge was incomplete. ALBO MPCA-II showed complete closure of the pulp opening by dentin bridge in 4 samples, while in the same number of teeth it was incomplete. In the control group, 4 teeth showed a complete dentin bridge, whereas in 6 teeth it was incomplete. Histological analysis indicated favourable therapeutic effects of Biodentine® and the two materials ALBO-MPCA I and ALBO-MPCA II after teeth pulp capping in Vietnamese pigs. Pulp reaction was similar to that caused by ProRoot MTA®.
AB  - Cilj ovog rada je bio dase ispita efekat Biodentina i dva nova nanostrukturna materijala na bazi aktivnih cilikatnih cemenata na eksponiranu pulpu zuba vijentamskih svinja. Istraživanje je obuhvatilo 40 zuba i dve vijetnamske svinje starosti 24 meseca. Na svima zubima je urađena preparacije kaviteta V klase i malim okruglim svrdlom eksponirana komora pulpe. Na perforaciju pulpe aplikovani su materijali: Biodentin (10 zuba), ALBO-MPCA I (10 zuba), ALBO-MPCA II (10 zuba). U kontrolnoj grupi pulpa je prekrivena MTA-om (10 zuba). Nakon observacionog perioda od 28 dana, životinje su žrtvovane i pripremljene za histološku analizu. Svetlosnom mikroskopijom analizirano je formiranje dentinskog mostića, postojanje reorganizacije tkiva, prisustvo inflamacije i bakterija u pulpi. Kod Biodentina kompletan dentinski mostić je zabeležen u 3 slučaja, a inkompletan dentinski mostić u vidu dentinskih ostrvaca u 4 slučaja. Nanostrukturni materijal ALBO-MPCA I je doveo do stvaranja kompletnog dentinskog mostića kod 5 zuba, a kod 3 zuba dentinski mostić je bio inkompletan. ALBO-MPCA II je pokazao kompletno zatvaranje komore pulpe dentinskim mostićem u 4 uzorka, dok je kod 4 uzorka on bio inkompletan. Kontrolni materijal MTA je kod 4 zuba imao stvoren kompletan dentinski mosti}, a u kod 6 zuba je on bio inkompletan.
