Expression and role of p53 in oral lichen planus patients

AB  - Purpose: Oral lichen planus (OLP) is an autoimmune skin and mucosal disorder. The range of malignant transformation in OLP varies between 0.1-3%. p53 is a tumor suppressor protein. Defective p53 could allow abnormal cells to proliferate, resulting in cancer. p53 plays an important role in cell cycle control and apoptosis and loss of p53 function has been demonstrated in about half of all human cancers. The purpose of the study was to investigate the malignant potential of OLP on the basis of p53 expression and to correlate p53 expression with clinical and histopathological features of OLP. Methods: 40 patients with OLP underwent biopsy. All tissue samples were treated immunohistochemicaly using avidin-biotin peroxidase complex method. Results: In 80% of OLP specimens the nuclei of basal and parabasal keratinocytes were p53-positive, but in low numbers. Low percentage of p53-positive cells in older and medium percentage of p53-positive cells in younger group of OLP patients were noted. Higher intensity of p53 stained keratinocytes, no matter their low number, could represent mutant and more stable form of p53 protein, and at the same time signal for monitoring of disease due to potential malignant transformation. Low percentage and weak intensity of p53-positive cells was detected mostly in OLP specimens with highly expressed civatte bodies (CB). Upregulation of apoptosis didn't correspond with the expression of CB. Conclusion: We believe that low percentage of p53-positive and well-marked keratinocytes in OLP represent the influence of mutant p53 protein, and that increasing expression of this protein could serve as a valuable diagnostic sign of early carcinogenesis. According to our results intensity of p53 coloration of keratinocytes could help assessing the malignant potential of OLP.

Changes in soft tissue profile following the treatment using a Herbst appliance: A photographic analysis

AB  - Background/Aim. Aesthetics is the reason for most of the class II malocclusion patients to opt for orthodontic treatment. In order to regulate retrognathic profile Herbst appliance for anterior movement of the mandible can be a treatment of choice. The aim of this study was to determine the soft tissue profile changes following Herbst appliance therapy on before and after treatment photos, using the computer program. Methods. This investigation was performed on profile photographs of 20 class II patients (12 females and 8 males) aged 18-23 years. Analysis of the changes in soft tissue facial structure relationships evident on the photographs before and after the Herbst appliance therapy was performed using Bentley Micro Station program. The first contour of the soft tissue profile was marked. The following reference lines were subsequently traced: Ricketts aesthetic E line and Juanita line. The area enclosed by these two lines included the nose, upper and lower lip, chin and free space in front of the lips. Using the computer program, the surfaces of the soft tissue structures and free space defined by the reference lines and profile contours were measured. Calculation of the relative proportion of surfaces was done for each photograph. The data obtained were then compared for each patient before and after the treatment. Skeletal and dentoalveolar treatment effects that support soft tissue changes were presented by the profile cephalometric parameters of sagital occlusion (SO) analysis. Results. A reduction in the relative surface of the upper lip in males (p  lt  0.01) and females (p  lt  0.05) was shown by the pictures. The space occupied by the chin was reduced after the treatment for females only (p  lt  0.05). The relative surface of the nasal soft tissues, that was included in the reference space was increased (p  lt  0.01) in both genders. The relationship between the soft tissue and empty surface was changed in favor of the empty surface (p  lt  0.05) in females. No statistically significant differences were found between the males and the females, before or after the Herbst appliance therapy. The soft tissue changes were the consequence of skeletal and dentoalveolar treatment effects: upper incisors retrusion, lower incisor protrusion and forward movement of the lower jaw. Conclusions. The Herbst appliance therapy caused a significant improvement of the profile appearance. The extent of soft tissue changes that occur on its dentoalveolar and skeletal support is a complex issue.
AB  - Uvod/Cilj. Većina mladih sa malokluzijom klase II odlučuje se za ortodontski tretman iz estetskih razloga. Za ispravljanje retrognatog profila može se koristiti aparat za anteriorno pomeranje mandibule - Herbst aparat. Cilj istraživanja bio je da se utvrde promene mekotkivnog profila posle primene Herbst aparata. Metode. Ispitivanje je izvršeno na profilnim fotografijama 20 mladih ljudi (12 ženskog i 8 muškog pola) sa malokluzijom klase II, starosti od 18 do 23 godine. Analizirane su promene odnosa mekotkivnih struktura lica na fotografijama načinjenim pre i posle sprovedene terapije Herbst aparatom. Za tu analizu korišćen je Bently Micro Station program. Digitalizovana je granica kontura mekotkivnog profila i ucrtavane su referentne linije: Ricketts-ova estetska E linija i Juanita linija. Ove linije ograničavaju prostor u kome su smešteni nos, gornja i donja usna i brada, kao i slobodni prostor koji se nalazi ispred konture mekotkivnog profila. Kompjuterski su merene površine mekotkivnih struktura i slobodnog prostora definisane referentnim linijama i konturom profila. Unutar slike izračunati su relativni odnosi površina. Nakon toga dobijeni podaci su poređeni pre i posle terapije za svaku osobu. Skeletni i dentoalveolarni terapijski efekti koji prouzrokuju mekotkivne promene prikazani su analizom sagitalne okluzije (SO) profilnog telerendgena. Rezultati. Sprovedenim merenjima utvrđeno je da je došlo do smanjenja relativne površine gornje usne kako kod dečaka (p  lt  0,01), tako i kod devojčica (p  lt  0,05). Prostor koji zauzima brada posle terapije značajno se smanjio samo kod devojčica (p  lt  0.05). Relativna površina mekotkivnog profila nosa obuhvaćena referentnim prostorom povećana je sa podjednakom statističkom značajnošću od p  lt  0,01 kod oba pola. Odnos mekog tkiva i praznog prostora promenio se u korist praznog prostora (p  lt  0,05) kod devojčica. Nije bilo statistički značajne razlike između dečaka i devojčica pre i posle sprovedene terapije. Retruzija gornjih i protruzija donjih sekutića, kao i mezijalno pomeranje donje vilice glavni su terapijski efekti koji izazivaju mekotkivne promene donje trećine lica. Zaključak. Terapija Herbst aparatom dovodi do značajnog poboljšanja izgleda profila lica. Prilagodljivost mekog tkiva na promene njegove koštane i dentoalveolarne potpore je kompleksna.

AB  - The aim of this study was to assess whether polymorphisms in toll-like receptor (TLR) and cluster of differentiation 14 (CD14) genes are associated with oral lichen planus (OLP) risk and clinical course of the disease. The study group consisted of 101 patients with confirmed OLP and 104 healthy blood donors without systemic or oral mucosal diseases. Single nucleotide polymorphisms of TLR2 (rs3804099), TLR3 (rs3775291 and rs5743312), TLR4 (rs4986790 and rs4986791), and CD14 (rs2569190) genes were genotyped using real-time PCR or PCR-restriction fragment length polymorphism (PCR-RFLP). The rs5743312 TLR3 gene polymorphism was associated with increased OLP risk in comparison with the wild type genotype (OR = 15.984, P = 0.011). No association with OLP risk was observed for the polymorphisms studied in TLR2, TLR4 and CD14 genes or for the rs3775291 polymorphism of the TLR3 gene. The polymorphisms of the TLR3 gene were in linkage disequilibrium (D ' = 1, r(2) = 0.1). Identified haplotypes were not associated with the risk of OLP. The findings of the current study suggest that the TT genotype of the rs5743312 TLR3 gene polymorphism may play a significant role in the aetiology of OLP.

abstract = "The aim of this study was to assess whether polymorphisms in toll-like receptor (TLR) and cluster of differentiation 14 (CD14) genes are associated with oral lichen planus (OLP) risk and clinical course of the disease. The study group consisted of 101 patients with confirmed OLP and 104 healthy blood donors without systemic or oral mucosal diseases. Single nucleotide polymorphisms of TLR2 (rs3804099), TLR3 (rs3775291 and rs5743312), TLR4 (rs4986790 and rs4986791), and CD14 (rs2569190) genes were genotyped using real-time PCR or PCR-restriction fragment length polymorphism (PCR-RFLP). The rs5743312 TLR3 gene polymorphism was associated with increased OLP risk in comparison with the wild type genotype (OR = 15.984, P = 0.011). No association with OLP risk was observed for the polymorphisms studied in TLR2, TLR4 and CD14 genes or for the rs3775291 polymorphism of the TLR3 gene. The polymorphisms of the TLR3 gene were in linkage disequilibrium (D ' = 1, r(2) = 0.1). Identified haplotypes were not associated with the risk of OLP. The findings of the current study suggest that the TT genotype of the rs5743312 TLR3 gene polymorphism may play a significant role in the aetiology of OLP.",

AB  - Purpose: The monoclonal antibody Ki-67 detects a nuclear antigen that is present only in proliferating cells. This is of particular interest for the analysis of the proliferation rates of malignant tumors. The aim of this study was to investigate the malignant potential of oral lichen planus (OLP) on the basis of expression of Ki-67 in healthy individuals (HI), patients with OLP and patients with squamous cell carcinoma (SCC), and to see for any potential interdependence between Ki-67 expression and different clinical and histopathological parameters in OLP. Methods: Immunohistochemistry for Ki-67 was carried out using an avidin-biotin peroxidase complex method. Results: Ki-67 was more expressed in keratinocytes and lymphocytes of OLP patients compared with HI, but less compared with patients with SCC. Keratinocytes and lymphocytes stained with Ki-67 in OLP patients were significantly higher in males, and in OLP specimens showed less developed civatte bodies (CB) and thickening of the basal membrane (TBM). Conclusion: Ki-67 may not serve as prognostic biomarker in oral cancer development from the initially diagnosed OLP, but it could help selecting patients with higher need of follow up for prevention of malignancy.

AB  - Background/Aim. Periodontitis is a chronic inflammatory disease of periodontal tissues with consequential is bone loss as a result of host immunological reactions caused by periopathogens. The aim of the study was to investigate if there is a correlation between clinical parameters and the presence of two most aggressive periopathogens (Aggregatibacter actinomycetemcomitans - Aa and Porphyromonas gingivalis - Pg) in patients with progressive periodontal lesions. Methods. A total of 34 systemic healthy people, 23 to 70 years old, were included in the study. The patients were clinically and radiologically examined, and after that, the representative pocket with greatest pocket depth was chosen and the sample was collected from that place. The measured clinic parameters were: gingival index, index of gingival bleeding, pocket depth and plaque indices. The multiplex Polymerase Chain Reaction (PCR) method was used for detection of periopathogens. After obtaining results, appropriate statistical tests were used to correlate the clinical and microbiological results. Results. Aa and Pg were detected in the same percentage of samples. Aa and Pg were detected in 35.29% samples alone, and in 29.41% both were detected. The values of measured clinical parameters did not show a statistical significance between the groups. In analysis of correlations among clinical parameters inside the groups, a statistical significance was found only between gingival and plaque index in the group with Aa. Conclusion. Clinical course of periodontitis in the developed stage does not differ in relation to the presence of different periopathogens as the major inductors of immunologically guided destructive processes.

Parodontopatija je hronično inflamatorno oboljenje parodontalnih tkiva koje za krajnji ishod ima gubitak potpornog koštanog tkiva zuba usled imunoloških reakcija izazvanih parodontopatogenim bakterijama. Cilj studije bio je korelisanje kliničkih parametara i prisustva dve najagresivnije parodontopatogene bakterije (Aggregatibacter actinomycetemcomitans - A.a. i Porphyromonas gingivalis - P.g.) kod bolesnika sa progresivnim parodontalnim lezijama. Metode. U studiju su bila uključena 34 sistemski zdrava ispitanika, starost 23-70 godina. Ispitanici su klinički i radiološki pregledani i uzorak je uziman iz reprezentativnog parodontalnog džepa sa najvećom dubinom sondiranja. Od kliničkih parametara mereni su gingivalni indeks, indeks krvarenja gingive, dubina parodontalnog džepa i indeks plaka. Prisustvo parodontopatogena dokazivano je multipleks metodom PCR (Polymerase Chain Reaction), a rezultati su korelisani sa kliničkim parametrima primenom odgovarajućih statističkih testova. Rezultati. Ista procentualna zastupljenost oba mikroorganizma dokazana je u uzorcima, naime i A.a. i P.g. bili su prisutni u po 35,39% uzoraka, a u 29,41% dokazana su oba mikroorganizma. Rezultati su korelisani po grupama formiranim u odnosu na prisustvo bakterija. Vrednosti merenih kliničkih parametara nisu se statistički značajno razlikovale u zavisnosti od prisustva parodontopatogena. Međusobne korelacije kliničkih parametara unutar grupe nisu pokazale statističku značajnost, osim korelacije gingivalnog i plak indeksa u grupi sa A.a. Zaključak. Klinički tok uznapredovale faze parodontopatije ne razlikuje se u odnosu na vrstu parodontalnih bakterija kao induktora imunološki posredovanih destruktivnih procesa.

AB  - Periodontal disease is a chronic, multi-factorial disease of the tissues supporting the teeth. Periodontitis in companion animals is an almost identical disease to that in humans in terms of disease course and clinical presentation. Receptor activator of NF-kB ligand (RANKL) and osteoprotegerin (OPG) are bioactive molecules that control bone resorption. This study aims to evaluate the effect of Chlorhexidine (CXD) on the RANKL and OPG expressions in gingival crevicular fluid (GCF) collected from subjects with chronic periodontitis. GCF was obtained from subjects with chronic periodontitis.10 subjects (CXD1) rinsed the mouth with 0.12% CXD, 10 subjects (CXD2) utilized 0.20% CXD and the last 10 (PL) used Placebo solution for 7 days. RANKL and OPG concentrations in GCF were measured by enzyme-linked immunosorbent assays ELISA at baseline and after 7 days. Periodontal clinical variables: clinical attachment loss (CAL), probing pocket depth (PPD), papilla-bleeding index (PBI) were evaluated in all groups. After 7 days in CXD1 and CXD2 group RANKL/OPG ratio exhibited a significant decrease (p lt 0.05) in contrast to the PL group where results showed similar values of RANKL/OPG ratio at baseline and after the observation period. RANKL/OPG ratio was positively correlated with PPD, CAL and PBI before and after the observation period in both Chlorhexidine (CXD1, CXD2) groups. In an existing inflammatory response, chlorhexidine reduced the level of periodontal inflammation, which leads to reduction of RANKL/OPG relative ratio. Decrease of RANKL/OPG ratio will apparently induce maintenance of alveolar bone and slow down periodontal tissue breakdown.
AB  - Parodontopatije su hronična, multikauzalna oboljenja potpornog aparata zuba. Parodontalna oboljenja koja srećemo kod kućnih ljubimaca su prema toku i kliničkoj slici skoro identična onima koje se javljaju kod ljudi. RANKL i osteoprotegerin (OPG) su bioaktivni molekuli koji kontrolišu koštanu resorpciju. Cilj ove studije je evaluacija efekata hlorheksidina na ekspresiju RANKL-a i OPG-a u gingivalnoj tečnosti (GT) uzetoj od pacijenata sa hroničnom parodontopatijom. 10 pacijenata (CXD1) su ispirali usta sa 0.12% CXD, 10 pacijenata (CXD2) su koristili 0.20% CXD i poslednjih 10 pacijenata (PL) su koristili placebo rastvor 7 dana. RANKL i OPG koncentracije u GT su merene ELISA testom na početku i posle sedam dana. Parodontalni klinički parametri CAL, PPD i PBI su evaluirani u svim grupama. Posle 7 dana u CXD1 i CXD2 grupi RANKL/OPG odnos je pokazao signifikantno smanjenje (p lt 0.05) u poređenju sa PL grupom gde su zabaleženi slični rezultati na početku i nakon opservacionog perioda. RANKL/OPG odnos je pokazao pozitivnu korelaciju sa vrednostima PPD-a, CAL-a i PBI-a pre i nakon observacionog perioda u obe eksperimentalne grupe (CXD1, CXD2). U prisutnom inflamatornom odgovoru hlorheksidin je redukovao nivo inflamacije, što je uslovilo redukciju RANKL/OPG odnosa. Rezultati istraživanja dokazuju da koncentracija hlorheksidina ne utiče statistički značajno na smanjenje RANKL/OPG odnosa.

Decrease of RANKL/OPG ratio will apparently induce maintenance of alveolar bone and slow down periodontal tissue breakdown., Parodontopatije su hronična, multikauzalna oboljenja potpornog aparata zuba. Parodontalna oboljenja koja srećemo kod kućnih ljubimaca su prema toku i kliničkoj slici skoro identična onima koje se javljaju kod ljudi. RANKL i osteoprotegerin (OPG) su bioaktivni molekuli koji kontrolišu koštanu resorpciju. Cilj ove studije je evaluacija efekata hlorheksidina na ekspresiju RANKL-a i OPG-a u gingivalnoj tečnosti (GT) uzetoj od pacijenata sa hroničnom parodontopatijom. 10 pacijenata (CXD1) su ispirali usta sa 0.12% CXD, 10 pacijenata (CXD2) su koristili 0.20% CXD i poslednjih 10 pacijenata (PL) su koristili placebo rastvor 7 dana. RANKL i OPG koncentracije u GT su merene ELISA testom na početku i posle sedam dana. Parodontalni klinički parametri CAL, PPD i PBI su evaluirani u svim grupama. Posle 7 dana u CXD1 i CXD2 grupi RANKL/OPG odnos je pokazao signifikantno smanjenje (p lt 0.05) u poređenju sa PL grupom gde su zabaleženi slični rezultati na početku i nakon opservacionog perioda. RANKL/OPG odnos je pokazao pozitivnu korelaciju sa vrednostima PPD-a, CAL-a i PBI-a pre i nakon observacionog perioda u obe eksperimentalne grupe (CXD1, CXD2). U prisutnom inflamatornom odgovoru hlorheksidin je redukovao nivo inflamacije, što je uslovilo redukciju RANKL/OPG odnosa. Rezultati istraživanja dokazuju da koncentracija hlorheksidina ne utiče statistički značajno na smanjenje RANKL/OPG odnosa.",

Methods: Immunohistochemistry for Bcl-2 was carried out using an avidin-biotin peroxidase complex method. Tissue sections were treated with mouse monoclonal antibody against Bcl-2 (124, DAKO A/S, Denmark; dilution 1/100). Immunohistochemical parameters measured included total tissue area, total stained area and intensity of stain. Results: Keratinocytes were not Bcl-2 positive in H and were sparsely positive in OLP. The number of lymphocytes stained with Bcl-2 was significantly lower in Has compared to patients with SCC and OLP. Bcl-2 staining was weak to moderate in OLP, and moderate to intense in SCC. Bcl-2-positive lymphocytes were more expressed in older OLP patients (>55 years), and in OLP specimens with orthokeratinized epithelium, less developed acanthosis and highest grade of lymphocyte expression. Conclusion: Bcl-2 may not serve as a prognostic biomarker in oral cancer development from OLP, but it could help in selecting patients with higher need of follow up to prevent malignancy.

AB  - Purpose: Oral complications are frequent and troublesome symptoms for those undergoing chemotherapy for cancer Several antineoplastic agents are proved to have stomatotoxic potential, among them 5-fluorouracil (5-FU). The aim of the present study was to evaluate the oral status and patient experiences during chemotherapy with 5-FU for colorectal cancer Methods: Twenty-eight patients treated with 5-day 5-FU plus leucovorin entered this study. Positive data about oral symptoms were taken by anamnesis. Mucositis severity index, gingival index, plaque index, probing pocket depth and bleeding on probing have been used to assess oral mucosa and periodontal status of the patients. Patients were examined prior to chemotherapy and 14 days after the start of the chemotherapy cycle. Results: Mild to moderate subjective complaints concerning oral cavity were reported by 17.9% of patients before and 39.2% of patients after chemotherapy. Clinical examination revealed oral mucosa damage in 10.7% and 35.7% of patients, with mean mucositis score of 0.14 and 0.54 before and after chemotherapy, respectively. Although mean values of all periodontal indices were elevated after chemotherapy, only increase in gingival index was statistically significant (p=0.035). Mucositis was significantly correlated with oral pain (p=0.00), xerostomia (p=0.00), and plaque index (p=0.077), while the correlation between mucositis and the rest of the examined parameters was not significant. Conclusion: Oral complications were not highly expressed in this study Although 5-FU is considered to exert significant stomatotoxic effect, severe mucositis was far less common in this study compared to studies reported elsewhere.

Circulating immune complexes, immunoglobulin G, salivary proteins and salivary immunoglobulin A in patients with Sjögren's syndrome

AB  - Introduction. Sjögren's syndrome (SS) is a chronic autoimmune disorder, with its major clinical manifestations resulting from changes in exocrine glands. Objective The aim of this study was to evaluate serum concentrations of circulating immune complexes (CIC) and immunoglobulin G (IgG), and salivary proteins (SP) and salivary immunoglobulin A (sIgA) in 40 patients with SS, and to correlate these values among themselves, as well as with the unstimulated salivary flow rate (USFR) and the duration of disease. Methods. The total of 40 patients were included in this research. CIC was determined using the solution of polyethylene glycol and IgG with the standard procedure of radial immunodiffusion. SP was investigated by the method of Lowry and sIgA was separated from the whole saliva using the method of immune chromatography. Results. The values of most of the studied parameters exceeded the normal range in a high degree: CIC 72.5%, IgG 70%, SP 80%. The concentrations of CIC were significantly higher in the patients with the duration of disease less than 10 years. With the decrease of USFR, the concentration of sIgA and IgG were increased with statistical significance. Conclusion The increased prevalence of abnormal values of CIC, IgG and SP indicate that the patients with SS have developed a higher level of immune reactivity. These results could be useful in diagnosis and disease activity monitoring.
AB  - Uvod. Sjegrenov (Sjögren) sindrom (SS) je hronična autoimuna bolest koja nastaje usled patoloških promena u egzokrinim žlezdama. Cilj rada Cilj rada je bio da se koncentracije cirkulatornih imunih kompleksa (CIC) i imunoglobulina G (IgG) u serumu, kao i salivarnih proteina (SP) i salivarnog IgA (sIgA) u pljuvački uporede međusobno i u odnosu na stepen protoka nestimulisane pljuvačke (USFR) i trajanja bolesti. Metode rada. Istraživanje je obuhvatilo 40 bolesnika sa SS. CIC je bio određen korišćenjem rastvora polietilenglikola, a IgG standardnim postupkom radijalne imunodifuzije. SP su izolovani metodom po Loriju (Lowry), dok je sIgA izdvojen iz ukupne pljuvačke korišćenjem metode imune hromatografije. Rezultati. Koncentracije većine ispitanih parametara su u velikoj meri odstupale od njihovih normalnih vrednosti: CIC 72,5%, IgG 70%, SP 80%. Koncentracije CIC su bile statistički značajno veće kod bolesnika čija je bolest trajala manje od 10 godina. Sa smanjenjem USFR koncentracije sIgA i IgG su se povećavale sa statističkom značajnošću. Zaključak. Povećane koncentracije CIC, IgG i SP kod bolesnika sa SS upućuju na povišen nivo imunološke reaktivnosti. Podaci dobijeni u ovom istraživanju mogu biti korisni prilikom posmatranja aktivnosti i dinamike promena kod SS.

Expression of Fas/FasL in patients with oral lichen planus

Results: Only a low percentage of infiltrating lymphocytes and no keratinocytes were Fas-positive in OLP specimens. The highest percentage of Fas-staining keratinocytes in our survey was identified mostly in H and patients with well-differentiated SCC In most cases of SCC, OLP and H a high percentage of keratinocytes and lymphocytes were FasL-positive. FasL expression was negatively correlated with the degree of cell differentiation and apoptosis. Taking into consideration that all carcinomas in this survey were highly differentiated, it is not surprising that no statistically significant differences in FasL expression between H, OLP and SCC specimens were detected Conclusion: Downregulation of Fas expression in keratinocytes and lymphocytes of OLP specimens, together with upregulation of FasL, may serve as initial prognostic biomarker in oral cancer development.

The highest percentage of Fas-staining keratinocytes in our survey was identified mostly in H and patients with well-differentiated SCC In most cases of SCC, OLP and H a high percentage of keratinocytes and lymphocytes were FasL-positive. FasL expression was negatively correlated with the degree of cell differentiation and apoptosis. Taking into consideration that all carcinomas in this survey were highly differentiated, it is not surprising that no statistically significant differences in FasL expression between H, OLP and SCC specimens were detected Conclusion: Downregulation of Fas expression in keratinocytes and lymphocytes of OLP specimens, together with upregulation of FasL, may serve as initial prognostic biomarker in oral cancer development.",

Association of unstimulated salivary flow rate and labial salivary gland histopathology with serologic features of Sjögren’s syndrome

AB  - Sjögren’s syndrome (SS) is a chronic autoimmune disorder with its major clinical manifestation resulting from changes in exocrine glands. An autoimmune etiology of SS is supported by the presence of autoantibodies specific for nuclear anti Ro (SS-A) and anti La (SS-B) antigens and other antinuclear antibodies (ANA) in sera of SS patients. The aim of the study was to evaluate the frequency of positive values of anti SS-A, anti SS-B antibodies and ANA in serum of 40 SS patients, and to correlate the above mentioned parameters with unstimulated salivary flow rate (USFR) and labial salivary gland focus score (LFS). Total of 40 patients was included in this research, 23 patients with primary SS and 17 with secondary SS. ANA was detected by the method of indirect immunofluorescence, and anti Ro/SS-A and anti La/SS-B antibodies were quantified by sandwich (Elisa) assay. The positive values of ANA were identified in plasma of 32 examined SS patients (80%), anti SS-A (Ro-52 kDa, Ro-60 kDa) antibodies were positive by 22 patients (55%), and anti SS-B antibodies were positive in less then 50% of examined SS patients (40%). The most presented were the patients with medium expressed xerostomia. Significant correlation between the unstimulated salivary flow rate and the positive values of ANA, anti Ro/SS-A, and anti La/SS-B antibodies could not be noticed. With increasing severity of disease there was an increase of the number of lymphocyte foci and a progressive loss of acinar tissue. High level of anti Ro/SS-A antibodies was found more often in plasma of the SS patients in a case of progressive loss of acinar tissue of the salivary gland (terminal stage), with statistical significance. It was concluded that the increased prevalence of positive values of anti Ro/SS-A antibodies and ANA indicates that the patient with SS has developed higher level of autoimmune reactivity. Level of anti Ro/SS-A antibodies in the peripheral blood of the patient with SS could be a significant marker of the degree of salivary/tissue destruction and disease severity.
