Assessment of bone vascularity in the posterior maxilla during dental implant insertion by laser Doppler flowmetry.

AB  - Bone quality is an important factor that may determine implant success. In addition to the other parameters that define bone quality, vascularity plays a vital role in the process of osseointegration of dental implants. Laser Doppler flowmetry (LDF) is an appropriate method for assessment of tissue vascularity at the level of microcirculation. It is non-invasive, painless and well accepted by the patients. The results are expressed in Perfusion Units (PU). The aim of this study was to estimate bone vascularity in the human posterior maxilla during implant insertion using LDF. Nine patients, three females and six males, mean age 57.56 +/- 8.83 years, consecutively treated with 54 implants were enrolled in this study. Implants were inserted in the posterior maxilla, in positions of first premolar, second premolar and first molar bilaterally in each patient. After implant site preparation with a drill of diameter 2.8 mm, bone vascularity was measured using LDF. Mean LDF value for 54 osteotomy sites was 43.39 +/- 14.65 PU. Results of the present study showed there wasn't statistically significant difference in LDF values between implant site positions and genders. It was also revealed that the proximity from the apical part of the implant site to the maxillary sinus floor had no influence on LDF values. Therefore, bone vascularity in the posterior maxilla could be measured by LDF during implant insertion, and those results might be considered as a standard data for this part of the upper jaw.

Comparison of pulpal anesthesia and cardiovascular parameters with lidocaine with epinephrine and lidocaine with clonidine after maxillary infiltration in type 2 diabetic volunteers

AB  - The pulpal anesthetic and cardiovascular parameters obtained by 2 % lidocaine with epinephrine (LE; 1:80,000) or clonidine (LC; 15 mcg/ml) were studied in diabetes mellitus (DM) type 2 and healthy volunteers (72), after maxillary infiltration anesthesia. Onset and duration of pulpal anesthesia were measured by electric pulp tester; vasoconstrictive effect of used local anesthetic mixtures by laser Doppler flowmetry (LDF) through pulpal blood flow (PBF); systolic blood pressure (SBP), diastolic blood pressure (DBP), and heart rate (HR) were registered by electrocardiogram monitoring. Onset of pulpal anesthesia was shorter for LC than for LE in healthy, while it was not different in diabetic participants; duration of pulpal anesthesia was significantly longer in type 2 diabetic participants, regardless of used anesthetic mixture. Significant reduction of PBF with LE was observed during 45 min in healthy and 60 min in diabetic participants, while with LC such reduction was observed during 45 min in both groups. LE caused a significant increase of SBP in the 5th and 15th minutes in diabetic versus healthy participants, while LC decreased SBP from the 10th to 60th minutes in healthy versus diabetic participants. DM type 2 influences duration of maxillary infiltration anesthesia obtained with LE and LC, and systolic blood pressure during LE anesthesia. The obtained results provide elements for future protocols concerning intraoral local anesthesia in DM type 2 patients.

Reduced muscarinic parotid secretion is underlain by impaired NO signaling in diabetic rabbits

AB  - ObjectivesThe influence of experimental diabetes (alloxan, 100mgkg(-1)) was studied on rabbit parotid gland function. Material and MethodsCarbachol-induced parotid secretion in vivo, and in vitro quantification of inducible nitric oxide synthase (iNOS) mRNA expression, by real-time RT-PCR, and activity of superoxide dismutase (SOD) and total antioxidant capacity (TAC) in commercial colorimetric assays were measured in parotid glands of non-diabetic and diabetic rabbits. ResultsCarbachol-induced dose-dependent increase in parotid secretion significantly reduced in diabetic rabbits. Functional studies in the presence of muscarinic receptor and nitric oxide synthase (NOS) antagonists revealed that in M-3 receptor-mediated carbachol secretion, nitric oxide, deriving mainly from neuronal NOS (nNOS) in control, and iNOS in diabetic rabbits, was involved. Also, upregulation of iNOS mRNA expression and enhanced SOD activity and TAC were detected in diabetic glands. ConclusionsOur data suggest that decreased M-3 receptor-mediated parotid secretion in diabetic rabbits appears to be due to alterations in NO signaling, mainly due to iNOS induction, accompanied by elevated antioxidant response.

Levobupivacaine vs. bupivacaine for third molar surgery: quality of anaesthesia, postoperative analgesia and local vascular effects

AB  - The main purpose of this prospective, double-blind, randomized study was to evaluate anaesthetic parameters, postoperative analgesia and vasoactive properties of levobupivacaine and bupivacaine for lower third molar surgery. Sixty patients (ASA I) were scheduled for lower third molar surgery under inferior alveolar nerve block, lingual nerve block and buccal nerve block (mandibular nerve blocks) obtained with 3 ml of 0.5 % levobupivacaine and 3 ml of 0.5 % bupivacaine. Success rate, onset and duration of three nerve bocks were evaluated by electrical pulp testing, pinprick testing and signs of soft tissue anaesthesia (patient-reported numbness). Intensity of intraoperative anaesthesia and postoperative analgesia were measured with visual analogue scale (VAS) and numeric rating scale (NRS). The time of first postoperative pain reported and analgesic consumption were also recorded. The laser Doppler flowmetry was used for the measurement of the first premolar pulpal blood flow. There were no differences between levobupivacaine and bupivacaine regarding the success rate, onset and duration of mandibular nerve blocks as well as intensity of postoperative analgesia and analgesic consumption. Intensity of intraoperative anaesthesia and duration of postoperative analgesia were significantly higher in the levobupivacaine than in the bupivacaine group. Both anaesthetics showed similar biphasic vasoactive effect. Levobupivacaine 0.5 % achieved superiority over bupivacaine 0.5 % in the intensity of intraoperative anaesthesia and duration of postoperative analgesia for lower third molar surgery under the mandibular nerve blocks. It seems that the plain levobupivacaine (0.5 %) could be an effective alternative to plain bupivacaine (0.5 %) in those dental procedures which require profound bone and soft tissue anaesthesia.

The aim of this study was to find whether or not blood perfusion of the bone around the prepared implant sites influences subsequent implant stability.Methods:Patients with bilaterally edentulous mandible were treated with dental implants. Bone vascularity in implant sites was previously noted using Laser Doppler Flowmetry (LDF). Implant stability quotient (ISQ) was measured during follow-up period of 26th week. Statistical distribution and correlation between LDF and ISQ values were presented. Other variables (type of implant loading; sex and distance from the apical part of implant sites to the roof of the mandibular canal) were collected and correlated with LDF values.Results:The mean recorded LDF value was 53.05 perfusion unit. Eighteen implants were immediately loaded, and the other 18 were early loaded. In the group of early loaded implants, a statistically significant correlation between mean value of LDF and changing value of resonance frequency analysis (P  lt  0.05) was noted at 5th, 6th, 12th, and 26th weeks. Using Pearson coefficient of correlation, there was no statistically significant relationship between other variables and LDF values.Conclusion:LDF values of implant sites might determine future implant stability.

The objective of this study was to investigate the effects of estrogen serum level changes associated with the menstrual cycle and postmenopause on dental pulp blood flow (PBF) as well as its dynamics. Methods: Young women at the menstrual phase (low serum estrogen levels) and in the mid-cycle phase of the menstrual cycle (high serum estrogen levels) and postmenopausal women were enrolled in the study. PBF and its oscillations were measured by laser Doppler flowmetry and analyzed by using wavelet transform. Serum levels of estradiol-17 beta were measured by immunoassay. Results: PBFs of young women in the menstrual phase and postmenopausal women were mutually similar and significantly lower than those of young women in the mid-cycle period. With respect to the mid-cycle phase, relative amplitude and power were significantly increased in the interval 0.0095-0.02 Hz and decreased in the intervals 0.02-0.06 and 0.06-0.2 Hz in the menstrual phase. A significant decrease in 0.0095-0.02 Hz and increase in 0.02-0.06, 0.06-0.2, 0.2-0.6, and 0.6-1.6 Hz intervals were observed in postmenopause. Conclusions: The study has shown that the menstrual phase of the menstrual cycle and postmenopause have similar PBF decrease, but PBF oscillations are differently affected in the mid-cycle phase. Blood flow measurement by laser Doppler method in orofacial region

AB  - Laser Doppler is a noninvasive, objective, reproducible and painless method for measuring blood flow in tissue microcirculation. This method is based on the Doppler effect, the change in frequency of light reflecting from blood cells in motion. Light from helium-neon laser through optical fibers and probes is directed to the surface of the tissue in which the flow is measured. Light portion is reflected from the cells in motion and changes the frequency while another portion is reflected from the static tissue maintaining the same frequency as the initial light. The total reflected light, with changed and original frequency, reaches photo detector in the same probe where the emitter is and it is transformed into electrical impulse. In the orofacial region the laser Doppler method is used to examine blood flow in the mandible, teeth pulp and masticator muscles. A significant drawback of the laser Doppler method is its sensitivity to the ambient conditions during measuring and the fact that blood flow is measured in all blood vessels of examined microregion. Therefore, the circulation of isolated individual blood vessels can not be monitored. Laser Doppler method can give reliable indicators of blood flow in mouth tissue and method is acceptable for the patients.
AB  - Laser dopler floumetrija je neinvazivna, objektivna, reproduktibilna i bezbolna metoda merenja protoka krvi na nivou mikrocirkulacije tkiva. Metoda je zasnovana na promeni frekvencije svetlosti pri odbijanju od krvnih ćelija u pokretu, odnosno na Doplerovom efektu. Svetlost helijum- neonskog lasera se putem optičkih vlakana i sonde s emiterom usmerava na površinu tkiva u kojem se ispituje protok. Deo svetlosti se odbija od ćelija u pokretu, pri čemu menja frekvenciju, a drugi deo svetlosti se odbija od statičnih tkiva zadržavajući istu frekvenciju kao i upadna svetlost. Ukupna odbijena svetlost s promenjenom i nepromenjenom frekvencijom dospeva do fotodetektora, koji je u okviru iste sonde kao i emiter, i pretvara se u električni impuls. U orofacijalnoj regiji laser dopler metoda je korišćena za ispitivanje protoka krvi u mandibuli, protoka u pulpi humanih zuba i u mastikatornim mišićima. Značajan nedostatak ove metode je njena osetljivost na uticaje iz okoline, kao i činjenica da se protok krvi meri u svim krvnim sudovima ispitivane mikroregije, pa se ne može izolovano pratiti cirkulacija u pojedinačnim krvnim sudovima. Ovom metodom se mogu dobiti pouzdani pokazatelji protoka krvi u tkivima usne duplje, a pacijenti je dobro prihvataju.

Principles of the laser Doppler method for application in dentistry

AB  - Laser Doppler floumetar measure dynamic changes in blood flow to a non invasive way recording movement of blood cells in microcirculation. This method is based on the change in frequency of light with the refusal of blood cells in motion, a phenomenon known as the Doppler effect. Results obtained in orofacial region show that the laser Doppler is valid method for tooth pulp vitality testing, monitoring of tooth vitality after teeth or jaws trauma, monitoring reactions of oral tissues to locally and systemically applied drugs electrical or thermal stimulation as well as for monitoring the reaction of pulp to orthodontics and ortognatics procedures.
AB  - Laser dopler floumetrija meri dinamske promene u protoku krvi na ne invazivni način registrujući kretanje krvnih ćelija u mikrocirkulaciji. Ova metoda se zasniva na promeni frekvencije svetlosti pri odbijanju od krvnih ćelija u pokretu, fenomen poznat kao Doplerov efekat. Rezultati dobijeni istraživanjima u orofacijalnoj regiji pokazuju da je laser Dopler validna metoda za ispitivanje vitaliteta zubne pulpe, praćenje vitaliteta posle traume zuba ili vilica, praćenju reakcije oralnih tkiva na lokalno i sistemno primenjene lekove električnu ili termalnu stimulaciju kao i za praćenje reakcije pulpe na ortodontske i ortognatske procedure.

Determination of cortisol levels in plasma and saliva in women exposed to chronic stress

AB  - Introduction: It is known fact from the literature data that acute and chronic stress influence serum cortisol level. As with serum cortisol, the level of cortisol in saliva is also a reliable marker of stress. Material and method: this study was performed on 25 individuals, females, age 40-50 years. The participants were exposed to long term stress, held two jobs of 8 working hours, six days a weak, domestic work was not included. All of them were high educated, and none of them were on other medication therapy Aim: The Aim of this study was to evaluate possible differences in cortisol levels in stimulated and non stimulated saliva and serum and determine their association with long term stress. Results: Obtained results showed that cortisol level in saliva do not depend on saliva flow rate and increases in the afternoon period in individuals exposed to chronic stress. Correlation (r) between salivary cortisol and plasma cortisol is r= 0.91. Non-invasive sampling procedure suggests that saliva may be used for cortisol measurements in situations where blood sampling is difficult to perform.
AB  - Uvod: U literaturi je poznato da akutni i hronični stres menjaju nivo kortizola u krvi. Kao i u plazmi, nivo kortizola u salivi je pouzdan stres marker. Cilj: Cilj ovog rada je bio da se odredi moguća koncentracija razlika kortizola u stimulisanoj i ne stimulisanoj salivi i plazmi kod osoba izloženih hroničnom stresu i utvrdi njihova međusobna zavisnost. Subjekti i metode: U ispitivanje je uključeno 25 ispitanica ženskog pola, starosti 40 godina izloženih hroničnom stresu tokom poslednjih pet godina. Uzimani su uzorci nestimulisane i stimulisane pljuvačke i uzorci plazme u sredini radne nedelje i na polovini radnog vremena, a koncentracija kortizola je određivana ELISA - imunoeseo testom - Eclia. Rezultati: Dobijeni rezultati pokazuju da se nivo kortizola ne menja u odnosu na protok salive i da se povećava tokom popodneva kod osoba izloženih hroničnom stresu. Korelacija (r) između nivoa kortizola u salivi i plazmi iznosi r = 0,91. Ne invazivna procedura omogućava da se saliva može koristiti za određivanje nivoa kortizola u situacijama kada je otežano uzimanje uzorka krvi.

Correlation (r) between salivary cortisol and plasma cortisol is r= 0.91. Non-invasive sampling procedure suggests that saliva may be used for cortisol measurements in situations where blood sampling is difficult to perform., Uvod: U literaturi je poznato da akutni i hronični stres menjaju nivo kortizola u krvi. Kao i u plazmi, nivo kortizola u salivi je pouzdan stres marker. Cilj: Cilj ovog rada je bio da se odredi moguća koncentracija razlika kortizola u stimulisanoj i ne stimulisanoj salivi i plazmi kod osoba izloženih hroničnom stresu i utvrdi njihova međusobna zavisnost. Subjekti i metode: U ispitivanje je uključeno 25 ispitanica ženskog pola, starosti 40 godina izloženih hroničnom stresu tokom poslednjih pet godina. Uzimani su uzorci nestimulisane i stimulisane pljuvačke i uzorci plazme u sredini radne nedelje i na polovini radnog vremena, a koncentracija kortizola je određivana ELISA - imunoeseo testom - Eclia. Rezultati: Dobijeni rezultati pokazuju da se nivo kortizola ne menja u odnosu na protok salive i da se povećava tokom popodneva kod osoba izloženih hroničnom stresu. Korelacija (r) između nivoa kortizola u salivi i plazmi iznosi r = 0,91. Ne invazivna procedura omogućava da se saliva može koristiti za određivanje nivoa kortizola u situacijama kada je otežano uzimanje uzorka krvi.",

AB  - The aim of the present study was to investigate the effects of caffeic acid phenethyl ester (CAPE) on wound healing capacity and repair of the pulp dentin complex on an experimental model. Eight young male pigs were used in this study. Arteficially preformed pulp lesions were treated either with calcium hydroxide (control group) or with CAPE powder (experimental group). The material for histological analysis was 4 and 14 weeks after pulp capping. Results of the histological analysis revealed that inflammatory reactions occurred in both groups. In the control group inflammation could be scored as mild to moderate, with a tendency to decrease during the observed period, while in the experimental group it was scored as severe in most of the specimens. Dentin bridge formation and complete reparation of the dentin-pulp complex was observed in most of the specimens of the control group, while the repair process in the experimental group was characterized by fibrous tissue proliferation. In the experimental group severe haemorrhagia, tissue oedema and dentin resorption were also observed. Based on reported results it could be concluded that capping material containing CAPE could not enhance the repair of the dentin-pulp complex, and healing of the dentin-pulp complex was characterized by scar tissue formation, rather than dentin bridge formation.
AB  - Cilj rad je bio da se ispita efekat fenetil estra kafeinske kiseline na proces zarastanja rane i reparaciju dentinsko-pulpnog kompleksa. Istraživanje je izvršeno na 8 svinja muškog pola t.m. 60-65 kg. Njima je arteficijelno napravljena lezija u dentinsko-pulpnom kompleksu, koja je tretirana kalcijum hidroksidom (kontrolna grupa) ili fenetil estrom kafeinske kiseline (eksperimentalna grupa). Materijal za histološku analizu uzet je 4 i 14 nedelja nakon prekrivanja pulpe. Rezultati histološ ke analize su ukazali da se u obe grupe uzoraka pojavila zapaljenska reakcija. U kontrolnoj grupi, ona je ocenjena kao blaga do umerena, sa tendencijom smanjivanja tokom observacionog perioda, dok je u eksperimentalnoj grupi, u najvećem broju uzoraka ona ocenjena kao teška. Kompletna reparacija dentinsko- pulpnog kompleksa sa stvaranjem dentinskog mostića uočena je samo u kontrolnoj grupi, dok je u eksperimentalnoj, zarastanje bilo karakterisano vezivno tkivnom proliferacijom. U velikom broju uzoraka eksperimentalne grupe uočeni su znaci krvarenja, edema i resorpcije dentina. Na osnovu rezultata dobijenih u ovoj studiji može se reći da fenetil estar kafeinske kiseline ne podstiče reparativne procese u dentinsko-pulpnom kompleksu, tako da proces zarastanja rane nije bio praćen formiranjem reparativnog dentina već vezivno-tkivnog ožiljka.

AB  - This study was conducted to observe the response of dog's dental pulp to hydrpxylapatite (Hap) and calcium hydroxide when used as pulp capping materials. After the pulps of 22 teeth were exposed, they were capped with either Hap or calcium hydroxide. Histological analysis was performed 8 weeks after treatment. Results showed that pulp exposure caused irreversible injury of odontoblasts, which were subsequently replaced by similar, odontoblast-like cells. These cells were able for reparative dentin bridge formation in 6 cases treated with calcium hydroxide and almost all the cases treated with hydroxylapatite. The presence of odontoblast-like cells seemed to be crucial for reparative dentinogenesis. The capping material was of less importance. In conclusion neither calcium hydroxide nor hydrxylapatite had completely satisfied all the requirements of an ideal capping material, although Hap can be recognized as a superior alternative to calcium hydroxide.
AB  - Cilj ovog rada je bio ispitivanje odgovora zubne pulpe pasa nakon njenog direktnog prekrivanja hidroksilapatitom (Hap) ili kalcijum hidroksidom. U eksperimentu su korišćena 22 zuba kod kojih je nakon arteficijelnog otvaranja, zubna pulpa prekrivana Hap-om ili kalcijum-hidroksidom. Osam nedelja nakon prekrivanja, urađena je histološka analiza. Dobijeni rezultati su ukazali da je tokom arteficijelnog otvaranja pulpe došlo do ireverzibilnog oštećenja odontoblasta i njihove zamene ćelijama sličnim odontoblastima. Odontoblastima-slične ćelije stvorile su mostić reparativnog dentina u 6 uzoraka iz grupe tretirane kalcijum hidroksidom i skoro svim uzorcima tretiranih Hap-om. Uočeno je da je prisustvo odontoblastima- sličnih ćelija presudno za uspeh reparativne dentinogeneze, dok je vrsta korišćenog materijala za direktno prekrivanje, bila od manje važnosti. Pri tome ni hidroksilapatit ni kalcijum hidroksid ne zadovoljavaju sve zahteve koji se postavljaju pred idealne materijale za direktno punjenje. Hap ipak predstavlja superiorniju alternativu u odnosu na kalcijum hidroksid.

AB  - Objective: Acute and chronic actions of lithium on salivation induced by agonists associated with receptor-linked hydrolysis of membrane inositol phospholipids (carbachol and phenylephrine) and by agonist linked to activation of adenylate cyclase (isoproterenol) were investigated. 
Material and methods: In anaesthetized rats, submandibular salivation induced by intravenous injection of carbachol, phenylephrine and isoproterenol, was measured and expressed as volume of fluid (microl) elicited per 100 mg wet weight of each gland per minute. The experiments were repeated after acute and chronic treatment of lithium (7 mg kg(-1)). The results were analysed with unpaired t-test. 
Results: Chronic, but not acute lithium treatment significantly decreases carbachol- and phenylephrine-induced salivation while isoproterenol-induced salivation was not changed neither after acute nor after chronic administration of lithium. 
Conclusion: The results suggest that hyposalivation during chronic lithium therapy could be mediated by alterations in the phosphatidylinositol cycle and a consequent lack of inositol after agonist stimulation.

Histological evaluation of bone response to bioactive ceramics as graft material in rats

AB  - This experimental study was carried out on 35 Albino Wister rats Artificially created bone defects were grafted with bioactive ceramic materials or control material. Histological analysis was preformed 8 weeks and 6 months after the implantation. The results revealed that bone regeneration occurred in defects treated with bioactive ceramic materials while in the defects treated with the control material signs of tissue rejection were present. As stated before, it could be concluded that bioactive ceramics can be recommended as bone substitutes in orthopedic surgery and dentistry.
AB  - Eksperimentalna studija sprovedena je na 35 Albino Wistar pacova kod kojih su, veštački napravljeni defekti u butnoj kosti, popunjavani bioaktivnim keramičkim materijalima i kontrolnim materijalom. Histološka procena zarastanja defekta vršena je 8 nedelja i 6 meseci nakon implantacije. Rezultati histološke analize ukazali su da je u defektima koji su tretirani biokeramičkim materijalima ostvarena koštana regeneracija, dok je u grupi koja je tretirana roze voskom, kao kontrolnim materijalom, bila prisutna reakcija odbacivanja. Na osnovu dobijenih rezultata može se zaključiti da se ispitivani biokeramički materijali mogu koristiti za popunjavanje koštanih defekta u ortopedskoj hirurgiji i stomatologiji.

Evidence that ampicillin and streptomycin can suppress autotomic behavior in rats

AB  - Autotomy is a phenomenon of deafferentation of the rat limb by transection of the sciatic and saphenous nerves, inducing an automutilation of the limb as a response to painful stimuli generated from the amputation neuroma. Autotomy has been proposed as an experimental model for neuralgia pain. The present study was designed to investigate the influence of streptomycin, ampicillin and chloramphenicol applied either in the vicinity of the nerve stump or intramuscularly. The course of autotomy was followed during 8 weeks using the scoring system of Wall et al. (1979). It was shown that streptomycin and ampicillin can suppress autotomy when applied topically whereas systemic use of the investigated antibiotics was efficient only when streptomycin was injected.
AB  - Eksperiment je izveden na 60 Albino Wistar pacova kod kojih je izvršeno presecanje n. ischiadicus-a i n. saphenus-a što je za posledicu imalo pojavu fenomena autotomije. Ovaj fenomen predstavlja eksperimentalni model bola, a ispoljava se samoodgrizanjem delova denervisanog stopala eksperimentalne životinje. Većina autora smatra da supstance i lekovi koji sprečavaju autotomiju utiču i na supresiju bola. Imajući ovo u vidu, cilj naših istraživanja bio je da ispitivane antibiotike primenimo na eksperimentalnom modelu posle ispoljavanja fenomena autotomije, kako bi utvrdili njihovo dejstvo na supresiju ovog fenomena. U isto vreme moguće je razmatrati i eventualno korišćenje ovih lekova u terapiji bola. Rezultati naših istraživanja pokazuju da streptomicin primenjen lokalno i intramuskularno sprečava autotomno ponašanje kod pacova, dok ampicilin sprečava razvoj ovog fenomena primenjen lokalno. Mehanizam delovanja ovih antibiotika još uvek nije u potpunosti razjašnjen, ali to ne umanjuje njihovu eventualnu primenu u terapiji hroničnog bola.

Influence of the endothelium on the vasorelaxant response to acetylcholine and vasoactive intestinal polypeptide in the isolated rabbit facial artery

AB  - The aim was to examine the influence of the endothelium on acetylcholine (ACh) and vasoactive intestinal polypeptide (VIP) functional responses in the isolated glandular branch of rabbit facial artery precontracted with phenylephrine as well as the potential contribution of nitric oxide (NO) and prostanoids in the ACh- and VIP-induced effects. Acetylcholine caused endothelium-dependent and VIP endothelium-independent relaxations of facial artery. The effect of ACh was partly inhibited by NG-monomethyl-l-arginine (l-NMMA, a non-selective NO synthase inhibitor) or by indomethacin (a cyclooxygenase inhibitor) while being completely blocked after concomitant addition of l-NMMA and indomethacin. The relaxation of the facial artery caused by ACh was unaffected by 65 mm KCl. The VIP-induced vasodilation was potentiated by forskolin (an adenylate cyclase stimulator) and partly reduced by l-NMMA or S-methyl-l-thiocitrulline (l-SMTC, a neuronal NO synthase inhibitor), whereas it was unaffected by indomethacin. These results suggest that ACh effects on the rabbit facial artery are mediated through release of endothelium-derived NO and cyclooxygenase products, while the effect of VIP is most probably mediated by an increase of cyclic adenosine 3',5'-monophosphate (cAMP) in vascular smooth muscles and by VIP-induced release of NO from perivascular nerve fibers.

Influence of the endothelium on the vasorelaxant response to acetylcholine and vasoactive intestinal polypeptide in the isolated rabbit facial artery

AB  - The aim was to examine the influence of the endothelium on acetylcholine (ACh) and vasoactive intestinal polypeptide (VIP) functional responses in the isolated glandular branch of rabbit facial artery precontracted with phenylephrine as well as the potential contribution of nitric oxide (NO) and prostanoids in the ACh- and VIP-induced effects. Acetylcholine caused endothelium-dependent and VIP endothelium-independent relaxations of facial artery. The effect of ACh was partly inhibited by N-G -monomethyl-L-arginine (L-NMMA, a non-selective NO synthase inhibitor) or by indomethacin (a cyclooxygenase inhibitor) while being completely blocked after concomitant addition of L-NMMA and indomethacin. The relaxation of the facial artery caused by ACh was unaffected by 65 mm KCl. The VIP-induced vasodilation was potentiated by forskolin (an adenylate cyclase stimulator) and partly reduced by L-NMMA or S-methyl-L-thiocitrulline (L-SMTC, a neuronal NO synthase inhibitor), whereas it was unaffected by indomethacin. These results suggest that ACh effects on the rabbit facial artery are mediated through release of endothelium-derived NO and cyclooxygenase products, while the effect of VIP is most probably mediated by an increase of cyclic adenosine 3',5'-monophosphate (cAMP) in vascular smooth muscles and by VIP-induced release of NO from perivascular nerve fibers.

Substance P-induced oxygen consumption in rat major salivary glands

SP elicited the concentration-dependent increase of oxygen consumption in all 3 glands. The increase was inhibit able by spamide II, the selective NK1 receptor antagonist, hut not by atropine, the muscarinic receptor antagonist. Respiratory effect of SP required the presence of Ca2+ in the incubation medium and was blocked by ouabain. SP had no significant effect on acetylcholine resting release from all 3 salivary glands. The study indicates that SP activates selective NK1 receptors, which are located only postsynaptically, and produces Ca2+-dependent and oubain-sensitive oxygen consumption in rat major salivary glands.
