AB  - Objective: We aimed to investigate alteration in cellular signaling mediated by vascular endothelial growth factor (VEGF) and parameters of oxidative stress/nitric oxide generation, superoxide dismutase (SOD) and neuronal nitric oxide synthase (nNOS), underlying altered functional mechanical loading of TMJ (temporomandibular joint) during lateral mandibular deviation. Design: Thirty-eight 5-week-old male Wistar rats were divided into experimental group, which received acrylic resin appliance that shifted mandible to the left during closure, and control group. Computed tomography and histomorphometry were used for condyle analyses, while samples of condyle, synovial membrane and m. masseter were analyzed with enzyme-linked immunosorbent assay and spectrophotometry to determine VEGF and nNOS protein concentrations, and SOD activity. Results: Experimental group of rats developed smaller and asymmetrical mandibles. Less of new bone and cartilage formation and larger bone marrow cavities area were found in the experimental group. Higher VEGF expression in condyle and m. masseter as well as higher nNOS expression in m. masseter and synovial membrane were found in the experimental compared to the control group. Alteration of SOD activity was found in m. masseter and synovial membrane in the experimental group. Conclusions: Lateral mandibular deviation induces mandibular and condylar morphological changes as well as significant cellular signaling alterations in condyle, synovial membrane and masticatory muscle. Cellular VEGF protein overexpression and oxidative stress/nitric oxide disbalance could be the mechanisms underlying unbalanced functional TMJ loading due to mandibular deviation.

Physico-chemical and biological properties of dental calcium silicate cements - literature review

AB  - Dental cement materials have been developed with the aim to replace hard dental tissues. The first material used for pulp capping, root canal obturation, bifurcation perforation and apexification is calcium hydroxide (in 1920). A half century later, glass-ionomer cements began to suppress it as dentine substitutes. Finally, in the 1990s, calcium silicate (CS) material appeared in the dental research community as the most promising dentine substitute capable to adequately meet all clinical requirements. The aim of this paper is to present an overview of literature related to studies about CS materials taking into account their physical, chemical and biological properties and clinical applications. This review aims to discuss beneficial and adverse characteristics of CSs concerning interactions to the hard dentine and soft pulp/periodontal tissues. This review article deals with the literature data about currently commercially available CS concerning laboratory and clinical findings. 109 scientific articles were analyzed of which 62 references reported in vitro and 26 in vivo investigations while 21 references comprised reports, reviews and books dealing with both, in vitro and in vivo investigations. Although further data collection is necessary, CSs are promising materials that represent a gold standard for numerous dental clinical procedures.

Direct pulp capping with novel nanostructural materials based on calcium silicate systems and hydroxyapatite

AB  - Introduction Direct pulp capping is an important therapeutic method that has goal to provide formation of dentin bridge and healing process of the pulp. The aim of this study was to investigate the effects of new nanostructural materials based on calcium silicate systems and hydroxyapatite on exposed dental pulp in Vietnamese pigs. Material and Methods The study was conducted on 30 teeth of two Vietnamese pigs (Sus scrofa verus). On buccal surfaces of incisors, canines and first premolars, class V cavities were prepared with a small round bur and pulp horn was exposed. In the first experimental group (10 teeth) the perforation was covered with new nanostructural material based on calcium silicate systems (CS). In the second experimental group, the perforation was covered with compound of calcium silicate systems and hydroxyapatite (HA-CS) (10 teeth). In the control group, exposed pulp was covered with Pro Root MTA® (Dentsply Tulsa Dental, Johnson City, TN, USA) (10 teeth). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in all groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of new nanostructural materials based on calcium silicate systems and hydroxyapatite for direct pulp capping in teeth of Vietnamese pigs.
AB  - Uvod Direktno prekrivanje pulpe je važan terapijski postupak koji ima zadatak da obezbedi zatvaranje pulpne komore i omogući proces zarastanja. Cilj ovog rada je bio da se ispita efekat novih nanostrukturnih materijala na bazi kalcijum-silikatnih sistema i hidroksiapatita na eksponiranu pulpu zuba vijetnamskih svinja. Materijal i metode rada Istraživanje je sprovedeno na 30 zuba kod dve vijetnamske svinje (Sus scrofa verus). Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase, pri čemu je malim okruglim svrdlom napravljena namerna eksploracija komore pulpe. U prvoj eksperimentalnoj grupi (10 zuba) perforacija je prekrivana novim nanostrukturnim materijalom na bazi kalcijum-silikatnih sistema (CS). U drugoj eksperimentalnoj grupi (10 zuba) perforacija je prekrivana smešom kalcijum-silikatnog sistema i hidroksiapatita (HA-CS). U kontrolnoj grupi (10 zuba) perforacija je prekrivana sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Rezultati Na svim zubima eksperimentalnih grupa i kontrolne grupe zabeleženo je stvaranje dentinskog mostića. Zapaljenje pulpe je bilo blago do umereno i u eksperimentalnim i u kontrolnoj grupi. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo gram-pozitivnih bakterija u pulpi. Zaključak Histološka analiza je ukazala na povoljne terapijske efekte novih nanostrukturnih materijala na bazi kalcijum-silikatnih sistema i hidroksiapatita u direktnom prekrivanju pulpe zuba vijetnamskih svinja.

AB  - AimTo evaluate in vitro cytotoxicity and in vivo inflammatory response to new nanostructural materials based on active calcium silicate systems (CS) and hydroxyapatite (HA-CS). MethodologyCytotoxicity of eluates of new nanostructural noncommercial materials CS and HA-CS, and MTA (White MTA, Angelus((R)) Solucoes Odontologicas, Londrina, Brazil) as a control, were tested using the MTT assay on MRC-5 cells. Eluates of set materials were tested in 100% and 50% concentrations, 24h, 7days and 21days post-elution. The pH values were determined for undiluted eluates of set materials. Polyethylene tubes containing the test materials (CS, HA-CS, MTA) were implanted in subcutaneous tissue of Wistar rats. Histopathological examinations were conducted at 7, 15, 30 and 60days after the implantation. Data were statistically analyzed using three-way and one-way anova Tukey's post hoc test as well as Kruskall-Wallis test with Dunn's post hoc test at =0.05. ResultsAll materials significantly reduced cell viability; especially when undiluted eluates were used (P lt 0.001). After 24h elution, cell viability was 101.8%, 49.5 +/- 4.2% and 61 +/- 7.4%, for MTA, and HA-CS, respectively. However, CS and HA-CS were significantly less toxic than the control material MTA (P lt 0.05). Cytotoxicity could be at least partially attributed to pH kinetics over time. Dilution of eluates of all tested materials resulted in better cell survival. Histological effects of enamel matrix derivative on exposed dental pulp

AB  - Introduction Direct pulp capping procedure is a therapeutic application of a drug on exposed tooth pulp in order to ensure the closure of the pulp chamber and to allow the healing process to take place. Objective The aim of this study was to examine the histological effects of Emdogain® on exposed tooth pulp of a Vietnamese pig (Sus scrofa verus). Methods The study comprised 20 teeth of a Vietnamese pig. After class V preparation on the buccal surfaces of incisors, canines and first premolars, pulp was exposed. In the experimental group, the perforations were capped with Emdogain® (Straumann, Basel, Switzerland), while in the control group pulp capping was performed with MTA® (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass-ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). The observational period was 28 days, after which the animal was sacrificed and histological preparations were made. A light microscope was used to analyze dentin bridge formation, tissue reorganization and inflammation, and the presence of bacteria in the pulp. Results The formation of dentin bridge was observed in the experimental and control groups. Inflammation of the pulp was mild to moderate in both groups. Angiogenesis and many odontoblast-like cells, responsible for dentin bridge formation, were observed. Necrosis was not observed in any case, nor were bacteria present in the pulp. Conclusion Histological analysis indicated a favorable therapeutic effect of Emdogain® Gel in direct pulp capping of Vietnamese pigs. Pulp reaction was similar to that of MTA®.
AB  - Uvod Direktno prekrivanje pulpe je važan terapijski postupak primene leka na eksponiranu pulpu zuba, čiji je cilj da obezbedi zatvaranje pulpne komore i omogući proces zarastanja. Cilj rada Cilj ovog rada je bio da se histološki ispita efekat gleđnog emdogaina na eksponiranu pulpu zuba vijetnamske svinje (Sus scrofa domesticus). Metode rada Istraživanje je urađeno na 20 zuba vijetnamske svinje. Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase s eksponiranjem komora pulpe. U eksperimentalnoj grupi perforacija je prekrivana proizvodom Emdogain® (Straumann, Bazel, Švajcarska), a u kontrolnoj sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer-cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Period posmatranja trajao je 28 dana, a nakon žrtvovanja životinja, napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, zapaljenjska reakcija pulpe, reorganizacija pulpnog tkiva i postojanje bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe uočeno je stvaranje dentinskog mostića. Upala pulpe je bila blaga do umerena i u eksperimentalnoj i u kontrolnoj grupi. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo bakterija u pulpi. Zaključak Histološka analiza je ukazala na povoljne terapijske efekte emdogaina u direktnom prekrivanju pulpe zuba vijetnamskih svinja. Reakcija pulpe bila je slična onima koje je izazvao MTA.

Direct pulp capping using Biodentine

AB  - Introduction Direct pulp capping is therapeutic method of applying medication on exposed pulp in order to allow bridge formation and healing process. The aim of this study was to investigate the effect of Biodentine on exposed dental pulp of Vietnamese pigs. Material and Methods The study was conducted on 20 teeth of Vietnamese pigs (Sus scrofa domesticus). On buccal surfaces of incisors, canines and first premolars, class V cavities were prepared and pulp was exposed. In the experimental group (six incisors, two canines and two premolars) the perforation was covered with BiodentineR (Septodont, Saint-Maur-des-Fosses, France). In the control group, the perforation was covered with MTAR (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, an inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in both groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of Biodentine for direct pulp capping in teeth of Vietnamese pigs. Findings were similar with Biodentine and MTA.

Cilj ovog rada je bio da se ispita efekat biodentina na eksponiranu pulpu zuba vijetnamske svinje. Materijal i metode rada Istraživanje je izvršeno na 20 zuba vijetnamske svinje (Sus scrofa domesticus). Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase, pri čemu je napravljena namerna eksploracija komora pulpe. U eksperimentalnoj grupi (šest sekutića, dva očnjaka i dva premolara) perforacija je prekrivana preparatom Biodentine ® (Septodont, Sent Mor de Fos, Francuska). U kontrolnoj grupi perforacija je prekrivana sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer- cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Opservacioni period trajao je 28 dana. Nakon žrtvovanja životinja napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, inflamatorna reakcija pulpe, reorganizacija pulpnog tkiva i prisustvo bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe zabeleženo je stvaranje dentinskog mostića. Zapaljenje pulpe je bilo blago do umereno i u eksperimentalnoj i u kontrolnoj grupi uzoraka. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo Gram-pozitivnih bakterija u pulpi. Zaključak Histološka analiza je ukazala na povoljne terapijske efekte biodentina u direktnom prekrivanju pulpe zuba vijetnamskih svinja. Reakcija pulpe bila je slična onima koje je izazvao MTA.

In the control group, the perforation was covered with MTAR (Dentsply Tulsa Dental, Johnson City, TN, USA). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, an inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in both groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of Biodentine for direct pulp capping in teeth of Vietnamese pigs. Findings were similar with Biodentine and MTA., Uvod Direktno prekrivanje pulpe je terapijski postupak primene leka na eksponiranu pulpu zuba radi zatvaranja pulpne komore i omogućavanja procesa zarastanja. Cilj ovog rada je bio da se ispita efekat biodentina na eksponiranu pulpu zuba vijetnamske svinje. Materijal i metode rada Istraživanje je izvršeno na 20 zuba vijetnamske svinje (Sus scrofa domesticus). Na vestibularnim površinama sekutića, očnjaka i prvih premolara urađene su preparacije kaviteta V klase, pri čemu je napravljena namerna eksploracija komora pulpe. U eksperimentalnoj grupi (šest sekutića, dva očnjaka i dva premolara) perforacija je prekrivana preparatom Biodentine ® (Septodont, Sent Mor de Fos, Francuska). U kontrolnoj grupi perforacija je prekrivana sa MTA® (Dentsply Tulsa Dental, Džonson Siti, Tenesi, SAD). Svi kaviteti su restaurirani glasjonomer- cementom (GC Fuji VIII, GC Corporation, Tokio, Japan). Opservacioni period trajao je 28 dana. Nakon žrtvovanja životinja napravljeni su histološki preparati na kojima su analizirani postojanje dentinskog mostića, inflamatorna reakcija pulpe, reorganizacija pulpnog tkiva i prisustvo bakterija. Rezultati Na svim zubima eksperimentalne i kontrolne grupe zabeleženo je stvaranje dentinskog mostića. Zapaljenje pulpe je bilo blago do umereno i u eksperimentalnoj i u kontrolnoj grupi uzoraka. Uočeni su znaci neoangiogeneze i mnoštvo ćelija sličnih odontoblastima koje su odgovorne za stvaranje dentinskog mostića. Nekroza nije zabeležena ni u jednom slučaju, kao ni prisustvo Gram-pozitivnih bakterija u pulpi. Nanostructured material ALBO-MPCA I provided complete dentin bridge formation in 5 teeth, in 3 teeth the formed dentin bridge was incomplete. ALBO MPCA-II showed complete closure of the pulp opening by dentin bridge in 4 samples, while in the same number of teeth it was incomplete. In the control group, 4 teeth showed a complete dentin bridge, whereas in 6 teeth it was incomplete. Histological analysis indicated favourable therapeutic effects of Biodentine® and the two materials ALBO-MPCA I and ALBO-MPCA II after teeth pulp capping in Vietnamese pigs. Pulp reaction was similar to that caused by ProRoot MTA®.
AB  - Cilj ovog rada je bio dase ispita efekat Biodentina i dva nova nanostrukturna materijala na bazi aktivnih cilikatnih cemenata na eksponiranu pulpu zuba vijentamskih svinja. Istraživanje je obuhvatilo 40 zuba i dve vijetnamske svinje starosti 24 meseca. Na svima zubima je urađena preparacije kaviteta V klase i malim okruglim svrdlom eksponirana komora pulpe. Na perforaciju pulpe aplikovani su materijali: Biodentin (10 zuba), ALBO-MPCA I (10 zuba), ALBO-MPCA II (10 zuba). U kontrolnoj grupi pulpa je prekrivena MTA-om (10 zuba). Nakon observacionog perioda od 28 dana, životinje su žrtvovane i pripremljene za histološku analizu. Svetlosnom mikroskopijom analizirano je formiranje dentinskog mostića, postojanje reorganizacije tkiva, prisustvo inflamacije i bakterija u pulpi. Kod Biodentina kompletan dentinski mostić je zabeležen u 3 slučaja, a inkompletan dentinski mostić u vidu dentinskih ostrvaca u 4 slučaja. Nanostrukturni materijal ALBO-MPCA I je doveo do stvaranja kompletnog dentinskog mostića kod 5 zuba, a kod 3 zuba dentinski mostić je bio inkompletan. Histological and morphometric aspects of ridge preservation with a moldable, in situ hardening bone graft substitute

AB  - Biphasic calcium phosphates (BCP) are widely used in alveolar ridge regeneration as a porous scaffold for new bone formation. The aim of this case series was to evaluate the regenerative effect of the combination of BCP and polylactide-co-glycolide (PLGA) which can serve as a barrier membrane during bone regeneration. The study included five patients. Four months into the healing period, bone samples were collected for histological and morphometric analyses. The results of morphometric analysis showed that newly formed bone represented 32.2 ± 6.8% of the tissue, 31.9 ± 8.9% was occupied by residual graft and 35.9 ± 13.5% by soft tissue. Active osteogenesis was seen around the particles of the graft. The particles were occupied mostly by immature woven bone and connective tissue. The quality and quantity of newly formed bone, after the use of BCP/PLGA for ridge preservation, can be adequate for successful implant therapy after tooth extraction.

The effects of hydroxyapatite and platelet rich plasma on apexogenesis in monkeys

AB  - There are very few data about the effects of endogenous growth factors in vital pulp therapy, and still they are often controversial. This study was undertaken with the aim to evaluate the effects of platelet rich plasma (PRP) in conjugation with hydroxyapatite (HAP), as pulp capping materials, on root and periodontium formation. Eight young monkeys (Cercopithecus Aethiops) with permanent dentition and incomplete root formation were involved in this study. After pulpotomy, the pulp lesion was capped with calcium hydroxide (control), HAP (experimental) or HAP in conjugation with PRP (experimental). Six moths later, the animals were sacrificed, the tissue was removed en block, and prepared for histological analysis in a routine way. The results of histological analysis revealed that the healing process, characterized by dentin bridge formation, maintained morphological and functional integrity of dental pulp and complete formation of dental root and surrounding periodontium. The inflammatory reaction was scored as mild to moderate, in almost all samples in all groups, suggesting the biocompatibility of the used materials. Materials used in this study are convenient as capping agents, contributing in maintaining the integrity of the pulp tissue and facilitating root and periodontium formation. According to histological data it could be suggested that hydroxyapatite in conjugation with endogenous growth factors, represent a superior alternative to other materials used in this study.
AB  - Podaci iz literature o primeni faktora rasta u vitalnoj terapiji pulpe su oskudni i često kontraverzni. Cilj našeg rada je bio da se ispita uticaj plazme bogate trombocitima u kombinaciji sa hidroksiapatitom, kao materijala za direktno prekrivanje pulpe, na završetak rasta korena i formiranje okolnog parodoncijuma. Istraživanje je sprovedeno na osam mladih majmuna (Cercopithecus aethiops), sa stalnom denticijom i nezavršenim rastom korena. Nakon pulpotomije, pulpna lezija je prekrivena kalcijum hidroksidom (kontolna grupa), hidroksiapatitom (eksperimentalna grupa I) ili hidroksiapatitom u kombinaciji sa plazmom bogatom trombocitima (eksperimentalna grupa II). Šest meseci kasnije životinje su žrtvovane, a uzeto tkivo pripremljeno je za histološku analizu. Rezultati histološke analize ukazali su da je zarastanje bilo karakterisano stvaranjem mineralizovanog dentinskog mostića, uz očuvanje morfološkog i funkcionalnog integriteta pulpe i završetak rasta korena i okolnog parodoncijuma u uzorcima svih ispitivanih grupa. Zapaljenska reakcija je gotovo u svim ispitivnim uzorcima bila ocenjena kao blaga ili umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Na osnovu dobijenih rezultata može se reći da su svi primenjeni materijali pogodni za direktno prekrivanje pulpe, da doprinose očuvanju integriteta pulpe i ubrzavaju završetak rasta korena i formiranje apikalnog parodoncijuma, s tim što hidroksipatit u kombinaciji sa endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na druge materijale korišćene u ovoj studiji.

Materials used in this study are convenient as capping agents, contributing in maintaining the integrity of the pulp tissue and facilitating root and periodontium formation. According to histological data it could be suggested that hydroxyapatite in conjugation with endogenous growth factors, represent a superior alternative to other materials used in this study., Podaci iz literature o primeni faktora rasta u vitalnoj terapiji pulpe su oskudni i često kontraverzni. Cilj našeg rada je bio da se ispita uticaj plazme bogate trombocitima u kombinaciji sa hidroksiapatitom, kao materijala za direktno prekrivanje pulpe, na završetak rasta korena i formiranje okolnog parodoncijuma. Istraživanje je sprovedeno na osam mladih majmuna (Cercopithecus aethiops), sa stalnom denticijom i nezavršenim rastom korena. Nakon pulpotomije, pulpna lezija je prekrivena kalcijum hidroksidom (kontolna grupa), hidroksiapatitom (eksperimentalna grupa I) ili hidroksiapatitom u kombinaciji sa plazmom bogatom trombocitima (eksperimentalna grupa II). Šest meseci kasnije životinje su žrtvovane, a uzeto tkivo pripremljeno je za histološku analizu. Rezultati histološke analize ukazali su da je zarastanje bilo karakterisano stvaranjem mineralizovanog dentinskog mostića, uz očuvanje morfološkog i funkcionalnog integriteta pulpe i završetak rasta korena i okolnog parodoncijuma u uzorcima svih ispitivanih grupa. Zapaljenska reakcija je gotovo u svim ispitivnim uzorcima bila ocenjena kao blaga ili umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Na osnovu dobijenih rezultata može se reći da su svi primenjeni materijali pogodni za direktno prekrivanje pulpe, da doprinose očuvanju integriteta pulpe i ubrzavaju završetak rasta korena i formiranje apikalnog parodoncijuma, s tim što hidroksipatit u kombinaciji sa endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na druge materijale korišćene u ovoj studiji.",

Histological and immunohistochemical evaluations of rat soft tissue response to bioceramical implants

AB  - The purpose of this study was to evaluate the tissue response induced by bioceramical materials (BCM) implanted in rat subcutaneous tissue. Thirty two rats were assigned to 6 groups, as following: control group i.e. not treated; dental ceramic (DC) implants; alfa tricalcium phosphate synthetised at 22 MPa (αTCP22) and at 45 MPa (αTCP45); hydroxyapatite synthetised at 22 MPa (HAP22) and at 45 MPa (HAP45). Contra lateral sides of all test animals represented the blank control surgically incisied at the paravertebral sockets. The experimental groups received subcutaneous implants of the test BCM materials. Samples were collected after 14 days and processed histologically. Inflammatory reactions were noted microscopically. The inflammatory infiltrate consisted of lymphocytes, polimorphonuclears, macrophages and mastocytes. At same time there were signs of increased fibrosis around the graft places. In conclusion, the implant sides compared with surgical incision wounds induced a foreign body-type granuloma with subsequent fibrosis around the grafts. The presence of a capsule at the interface with BCM in rat subcutis did not elicit any remarkable immune responses, thus being considered biocompatible. By evaluating different responses according to BCM, the most intense tissue reaction was noticed at HAP45, graded as severe, and mild to moderate for DC, HAP22 and alfa tricaltium phosphate.
AB  - Svrha ove studije je bila procena tkivnog odgovora indukovanog biokeramič kim materijalima (BCM) inplantiranim u subkutano tkivo pacova. Trideset dva pacova je bilo podeljeno u sledećih 6 grupa: kontrolna grupa-neoperisana; inplantirane sa: dentalnom keramikom (DC); alfa trikalcijum fosfatima sintetisanim na 22 MPa (αTCP22) i na 45 MPa (αTCP45); hidroksiapatitom sintetisanim na 22 MPa (HAP22) i na 45 MPa (HAP45). Kontralateralne strane svih životinja bile su slepa kontrola i urađene su kao džepovi hirurške incizije u subkutanom tkivu. Eksperimentalne grupe su imale subkutano inplantirane biokeramičke materijale. Uzorci su sakupljeni posle 14 dana i sprovedeni u histološkoj proceduri, a inflamatorna reakcija je praćena mikroskopski. Inflamatorni infiltrat se sastojao od limfocita, polimorfonuklearnih leukocita, makrofaga i mastocita. Istovremeno su uočeni znaci rastuće fibroze oko mesta graftova. Histological evaluation of platelet rich plasma and hydroxiapatite in apexogenesis: Study on experimental animals

AB  - Background/Aim. There are very few data about the effects of endogenous growth factors in vital pulp therapy, and still they are often controversial. The aim of the study was to evaluate the effects of platelet rich plasma (PRP) in conjugation with hydroxyapatite (HAP), as pulp capping materials, to root and periodontium formation. Methods. Eight young monkeys (Cercopithecus Aethiops) with permanent dentition and incomplete root formation were involved in this study. After pulpotomy, the pulp lesion was capped with calcium hydroxide (control), hydroxyapatite (experimental group I) or hydroxyapatite in conjugation with PRP (experimental group II). Six months later, the animals were sacrificed, the tissue was removed en block, and prepared for the histological analysis in a routine way. Results. The results of the histological analysis revealed that healing process was characterised by dentin bridge formation, maintained morphological and functional integrity of dental pulp and complete formation of dental root and surrounding periodontium. The inflammatory reaction was scored as mild to moderate, in almost all the samples in all groups, suggesting the biocompatibility of the used materials. Conclusion. Materials used in this study are convenient as capping agents, contributing maintaining the integrity of the pulp tissue and facilitating root and periodontium formation. AB  - Uvod/Cilj. Podaci o primeni endogenih faktora rasta u terapiji vitalne pulpe zuba sa nezavršenim rastom korena su veoma oskudni i često kontraverzni. Cilj ovog rada bio je da se ispita uticaj plazme bogate trombocitima (PRP) u kombinaciji sa hidroksiapatitom (HAP), kao materijala za direktno prekrivanje pulpe, na završetak rasta korena i formiranje okolnog parodoncijuma. Metode. Istraživanje je obavljeno na osam mladih majmuna Cercopithecus Aethiops. Sve životinje imale su zube stalne denticije, sa nezavršenim rastom korena. Nakon trepanacije pulpnog prostora, pulpa je uklonjena do nivoa gleđnocementnog spoja, a lezija u pulpi prekrivena je kalcijum hidroksidom (kontrolna grupa), hidroksiapatitom (eksperimentna grupa I) i hidroksiapatitom u kombinaciji sa autogenim PRP faktorom (eksperimentna grupa II). Šest meseci nakon amputacije pulpe, životinje su žrtvovane, a tkivo za histološku analizu uzeto je u obliku blok sekcije i pripremljeno za mikroskopsku analizu. Rezultati. Zarastanje rane u pulpno-dentinskom kompleksu u obe eksperimentne i u kontrolnoj grupi bilo je karakterisano stvaranjem dentinskog mostića procesom reparativne dentinogeneze, očuvanjem morfološkog i funkcijskog integriteta pulpe, kao i završetkom rasta korena i stvaranjem okolnog parodoncijuma. U većini uzoraka obe grupe, zapaljenska reakcija ocenjena je kao blaga do umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Zaključak. Svi primenjeni materijali pogodni su za direktno prekrivanje pulpe, doprinose očuvanju njenog morfološkog i funkcionalnog integriteta i omogućavaju završetak rasta korena i formiranje okolnog parodoncijuma. Ipak, povoljniji rezultati dobijeni su u grupi koja je tretirana HAP/PRP, pa se može reći da HAP obogaćen endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na ostale materijale primenjene u ovoj studiji.

U većini uzoraka obe grupe, zapaljenska reakcija ocenjena je kao blaga do umerena, što govori u prilog biokompatibilnosti primenjenih materijala. Zaključak. Svi primenjeni materijali pogodni su za direktno prekrivanje pulpe, doprinose očuvanju njenog morfološkog i funkcionalnog integriteta i omogućavaju završetak rasta korena i formiranje okolnog parodoncijuma. Ipak, povoljniji rezultati dobijeni su u grupi koja je tretirana HAP/PRP, pa se može reći da HAP obogaćen endogenim faktorima rasta predstavlja superiorniju alternativu u odnosu na ostale materijale primenjene u ovoj studiji.",

Comparative immunohistochemical and quantitative analysis of inflammatory cells in symptomatic and asymptomatic chronic periapical lesions

It has been demonstrated that lymphocytes, plasma cells, macrophages and neutrophil granulocytes represent the predominant cells of the inflammatory lesion of the dental granulomas. Other cells, such as mast cells, eosinophils, dendritic cells comprise minor, but functionally important cell populations. Most of the data considering cells that take part in these processes have been derived from immunohistological studies. This study was undertaken with the aim to determine the phenotype profile of inflammatory cells of dental granulomas using immunohistochemical method in order to study the differences of their quantitative properties and distribution between symptomatic and asymptomatic lesions. Methods. The material for the analysis originated from 42 individuals with clinic and radiographic diagnosis of chronic periapical lesions. The tissue was take either during the periradicular surgery, or tooth extraction. Cryostat tissue sections were stained using the alkaline phosphatase-antialkaline phosphatase assay (APAAP). This method is highly valid and sensitive using a panel of specific monoclonal antibodies: CD3, CD4, CD8, CD19, CD38, CD14, CD1a, CD83, CD80, CD86, CD45 and CD123. Results. The composition of the cell population revealed that there was no homogenous and site-specific pattern of the distribution of inflammatory cells. The results of our investigation revealed that the majority of inflammatory cells comprised lymphocytes and plasma cells, followed by subpopulations CD4+, CD8+ and CD14+ cells. Much lower in number were CD80+, CD86+ and CD83+ and CD1a+ cells. There were no statistically significant differences in mean values of inflammatory cells number between symptomatic and asymptomatic lesions, with the exception of CD86+ cells, the number of which was statistically higher in symptomatic lesions. Conclusion. Inflammatory infiltrate cells in dental granulomas are dominated by T- and Blymphocytes. It points out the complexity of immunopathogenic events in imitiating and progressing of dental granulomas that involve mechanisms of both cellular and humoral immunity. Regarding the quantitative presence of immunocompetent cells in symptomatic and asymptomatic lesions no statistically significant difference was determined unless in mature dendritic cells present in symptomatic lesions.

Rezultati ranijih imunohistohemijskih studija pokazali su da su najbrojnije ćelije u hroničnim periapeksnim lezijama limfociti, plazmociti, makrofagi i neutrofilni granulociti, dok ostale ćelije, pre svega mastociti, eozinofilni granulociti i dendritske ćelije predstavljaju manje, ali funkcionalno značajne ćelijske populacije. Cilj ovog rada bio je da se korišćenjem relevantne imunohistohemijske metode determiniše fenotipski profil ćelija zapaljenskog infiltrata dentalnih granuloma, kao i da se odrede razlike u kvantitativnim odnosima i distribuciji ćelija između simptomatskih i asimptomatskih hroničnih periapeksnih lezija. Metode. Materijal za analizu dobijen je od 42 bolesnika sa kliničkom i radiografskom dijagnozom hronične periapeksne lezije. Tkivo je uzeto tokom periradikulne hirurške intervencije ili ekstrakcije zuba. Metoda primenjena na smrznutim isečcima bila je alkalna fosfatazaantialkalna fosfataza (APAAP). Rezultati analize pokazali su da nije postojalo homogeno područje zapaljenja, predodređeno da bude infiltrisano određenim tipom zapaljenskih ćelija. Utvrdili smo da su najbrojnije ćelije zapaljenskog infiltrata bili limfociti i plazmociti, zatim CD4+, CD8+ i CD14+, dok je broj CD80+, CD86+ i CD83+ i CD1a+ ćelija bio znatno manji. Srednje vrednosti broja zapaljenskih ćelija nisu pokazivale statistički značajne razlike između grupe simptomatskih i asimptomatskih periapeksnih lezija, osim CD86+ ćelija, čiji je broj bio statistički značajno veći u grupi simptomatskih lezija. Zaključak. Među ćelijama zapaljenskog infiltrata u dentalnim granulomima dominiraju T i B limfociti. To ukazuje na kompleksnost imunopatogenetskih zbivanja u inicijalnoj fazi i progresiji dentalnih granuloma u koje su uključeni mehanizmi celularnog i humoralnog imuniteta. U pogledu kvantitativne zastupljenosti imunokompetentnih ćelija u simptomatskim i asimptomatskim lezijama nije ustanovljena statistički značajna razlika, osim u pogledu zrelih dendritskih ćelija koje su bile zastupljenije kod simptomatskih lezija.

The effect of caffeic acid phenethyl ester on healing capacity and repair of the dentin-pulp complex: In vivo study

AB  - The aim of the present study was to investigate the effects of caffeic acid phenethyl ester (CAPE) on wound healing capacity and repair of the pulp dentin complex on an experimental model. Eight young male pigs were used in this study. Arteficially preformed pulp lesions were treated either with calcium hydroxide (control group) or with CAPE powder (experimental group). The material for histological analysis was 4 and 14 weeks after pulp capping. Results of the histological analysis revealed that inflammatory reactions occurred in both groups. In the control group inflammation could be scored as mild to moderate, with a tendency to decrease during the observed period, while in the experimental group it was scored as severe in most of the specimens. Dentin bridge formation and complete reparation of the dentin-pulp complex was observed in most of the specimens of the control group, while the repair process in the experimental group was characterized by fibrous tissue proliferation. In the experimental group severe haemorrhagia, tissue oedema and dentin resorption were also observed. Based on reported results it could be concluded that capping material containing CAPE could not enhance the repair of the dentin-pulp complex, and healing of the dentin-pulp complex was characterized by scar tissue formation, rather than dentin bridge formation.
AB  - Cilj rad je bio da se ispita efekat fenetil estra kafeinske kiseline na proces zarastanja rane i reparaciju dentinsko-pulpnog kompleksa. Istraživanje je izvršeno na 8 svinja muškog pola t.m. 60-65 kg. Njima je arteficijelno napravljena lezija u dentinsko-pulpnom kompleksu, koja je tretirana kalcijum hidroksidom (kontrolna grupa) ili fenetil estrom kafeinske kiseline (eksperimentalna grupa). Materijal za histološku analizu uzet je 4 i 14 nedelja nakon prekrivanja pulpe. Rezultati histološ ke analize su ukazali da se u obe grupe uzoraka pojavila zapaljenska reakcija. U kontrolnoj grupi, ona je ocenjena kao blaga do umerena, sa tendencijom smanjivanja tokom observacionog perioda, dok je u eksperimentalnoj grupi, u najvećem broju uzoraka ona ocenjena kao teška. Kompletna reparacija dentinsko- pulpnog kompleksa sa stvaranjem dentinskog mostića uočena je samo u kontrolnoj grupi, dok je u eksperimentalnoj, zarastanje bilo karakterisano vezivno tkivnom proliferacijom. U velikom broju uzoraka eksperimentalne grupe uočeni su znaci krvarenja, edema i resorpcije dentina. Na osnovu rezultata dobijenih u ovoj studiji može se reći da fenetil estar kafeinske kiseline ne podstiče reparativne procese u dentinsko-pulpnom kompleksu, tako da proces zarastanja rane nije bio praćen formiranjem reparativnog dentina već vezivno-tkivnog ožiljka.

Histological evaluation of odontoblast-like cells response after capping application of calcium hydroxide and hydroxilapatite in dog’s pulp

AB  - This study was conducted to observe the response of dog's dental pulp to hydrpxylapatite (Hap) and calcium hydroxide when used as pulp capping materials. After the pulps of 22 teeth were exposed, they were capped with either Hap or calcium hydroxide. Histological analysis was performed 8 weeks after treatment. Results showed that pulp exposure caused irreversible injury of odontoblasts, which were subsequently replaced by similar, odontoblast-like cells. These cells were able for reparative dentin bridge formation in 6 cases treated with calcium hydroxide and almost all the cases treated with hydroxylapatite. The presence of odontoblast-like cells seemed to be crucial for reparative dentinogenesis. The capping material was of less importance. In conclusion neither calcium hydroxide nor hydrxylapatite had completely satisfied all the requirements of an ideal capping material, although Hap can be recognized as a superior alternative to calcium hydroxide.
AB  - Cilj ovog rada je bio ispitivanje odgovora zubne pulpe pasa nakon njenog direktnog prekrivanja hidroksilapatitom (Hap) ili kalcijum hidroksidom. U eksperimentu su korišćena 22 zuba kod kojih je nakon arteficijelnog otvaranja, zubna pulpa prekrivana Hap-om ili kalcijum-hidroksidom. Osam nedelja nakon prekrivanja, urađena je histološka analiza. Dobijeni rezultati su ukazali da je tokom arteficijelnog otvaranja pulpe došlo do ireverzibilnog oštećenja odontoblasta i njihove zamene ćelijama sličnim odontoblastima. Odontoblastima-slične ćelije stvorile su mostić reparativnog dentina u 6 uzoraka iz grupe tretirane kalcijum hidroksidom i skoro svim uzorcima tretiranih Hap-om. Uočeno je da je prisustvo odontoblastima- sličnih ćelija presudno za uspeh reparativne dentinogeneze, dok je vrsta korišćenog materijala za direktno prekrivanje, bila od manje važnosti. Pri tome ni hidroksilapatit ni kalcijum hidroksid ne zadovoljavaju sve zahteve koji se postavljaju pred idealne materijale za direktno punjenje. Hap ipak predstavlja superiorniju alternativu u odnosu na kalcijum hidroksid.

Preimplant preparation of the extraction alveolus with the deproteinized bovine bone and calcium-sulphate

AB  - Background. Different materials are used to prevent the resorption of alveolar bone. The aim of this report was to show the radiographical and histological results prior to implant insertion, when a deproteinized bovine bone mineral (BioOss) and calcium-sulphate were placed into the extraction socket immediately after the tooth removal. Case report. A 22-year-old woman was scheduled for the removal of the second lower molar when the extraction socket was filled with BioOss covered with calcium-sulphate as a resorbable membrane. Primary closure of the surgical site was performed. Radiography was done 4 and 12 months later. One year after the surgery, when the implant was inserted, a biopsy of the new regenerated bone was obtained for the histological evaluation. The lamellar bone was evident using both materials. The resorption of BioOss was slow and the connective tissue was observed. Conclusion. Both materials had biocompatible and oseoconductive properties. One year after the grafting procedure, we observed the lamellar bone and partial resorption of BioOss, while calciumsulphate showed no significant effect as a resorbable membrane.
AB  - Uvod. Primenom različitih materijala sprečava se resorpcija koštanog tkiva na mestu izvađenog zuba. Cilj rada je bio da se prikažu rezultati radiografske i histološke analize posle kombinovane primene deproteinizovane goveđe kosti (BioOss) i kalcijum-sulfata postavljenih u alveolu neposredno posle vađenja zuba, a radi planirane ugradnje endosealnog implantata. Prikaz bolesnika. Posle vađenja donjeg drugog molara, kod bolesnice stare 22 godine, u alveolu je postavljen BioOss prekriven kalcijum-sulfatom u funkciji resorptivne membrane. Operativna regija je primarno ušivena. Kontrolni radiografski snimci urađeni su posle četvrtog i dvanaestog meseca. Godinu dana kasnije, neposredno pre ugradnje implantata, uzet je isečak novoformiranog koštanog tkiva za histološku analizu. Uočeno je stvaranje zrele lamelarne kosti, pri čemu je resorpcija BioOss bila usporena. Takođe, uočeno je i prisustvo vezivnog tkiva. Zaključak. Oba materijala su pokazala biokompatibilnost i oseokonduktivni efekat, stvaranje lamelarne kosti i delimičnu resorpciju BioOss-a godinu dana posle njihove primene. Prisutno vezivno tkivo je pokazalo da kalcijum-sulfat nije ispunio u potpunosti zahteve resorptivne membrane.

Characterization of antigen-presenting cells in human apical periodontitis lesions by flow cytometry and immunocytochemistry

AB  - Aim To analyse phenotypic characteristics of antigen-presenting cells (APC), isolated from human periapical lesions by flow cytometry and immunocytochemistry. Methodology Sixteen periapical lesions were digested for 15 min with 0.05% collagenase. Mononuclear cells, separated from other inflammatory cells by density centrifugation, were processed for flow cytometry and/or immunocytochemistry. Single and double immunostainings were performed using monoclonal antibodies specific for human CD45, CD3, CD19, CD14, HLA-DR, CD1a, CD83 and CD123. Results Antigen-presenting cells (HLA-DR+ cells) represented 32.9 +/- 17.8% of total mononuclear cells. Amongst them, B cells (HLA-DR+ CD19(+)) were the predominant APC population, followed by activated macrophages (HLA-DR+ CD14(+)), dendritic cells (DC) (HLA-DR+ CD14(-) CD19(-) CD3(-)) and activated T cells (HLA-DR+ CD3(+)). Based on the predominance of T cells (CD3(+)) or B cells and plasma cells (CD19(+) and CD19(lo), respectively) amongst mononuclear cell infiltrates, lesions were divided into T- and B-types. The percentage of DC in T-type lesions (27.1 +/- 6.8% of total HLA-DR+ cells) was higher, compared with B-type lesions (10.3 +/- 5.2%) (P  lt  0.01). Within the DC population, the percentages of CD1a (Langerhans cell type) and CD123 (probably plasmacytoid DC type) did not differ significantly between the groups (P > 0.05). However, the percentage of mature DC (CD83(+)) was significantly higher in T-type periapical lesions (P  lt  0.05). Conclusion Flow cytometry and immunocytochemistry are suitable methods for phenotypic analysis of APC after their isolation from human periapical lesions. APC, that were phenotypically heterogeneous, constituted a significant component of infiltrating cells. Lesions with the predominance of T cells were characterized by a higher proportion of mature DC (HLA-DR(+)CD83(+) cells) than lesions with predominance of B cells/plasma cells.

Histological evaluation of bone response to bioactive ceramics as graft material in rats

AB  - This experimental study was carried out on 35 Albino Wister rats Artificially created bone defects were grafted with bioactive ceramic materials or control material. Histological analysis was preformed 8 weeks and 6 months after the implantation. The results revealed that bone regeneration occurred in defects treated with bioactive ceramic materials while in the defects treated with the control material signs of tissue rejection were present. As stated before, it could be concluded that bioactive ceramics can be recommended as bone substitutes in orthopedic surgery and dentistry.
AB  - Eksperimentalna studija sprovedena je na 35 Albino Wistar pacova kod kojih su, veštački napravljeni defekti u butnoj kosti, popunjavani bioaktivnim keramičkim materijalima i kontrolnim materijalom. Histološka procena zarastanja defekta vršena je 8 nedelja i 6 meseci nakon implantacije. Rezultati histološke analize ukazali su da je u defektima koji su tretirani biokeramičkim materijalima ostvarena koštana regeneracija, dok je u grupi koja je tretirana roze voskom, kao kontrolnim materijalom, bila prisutna reakcija odbacivanja. Na osnovu dobijenih rezultata može se zaključiti da se ispitivani biokeramički materijali mogu koristiti za popunjavanje koštanih defekta u ortopedskoj hirurgiji i stomatologiji.

Rezultati histološke analize ukazali su da je u defektima koji su tretirani biokeramičkim materijalima ostvarena koštana regeneracija, dok je u grupi koja je tretirana roze voskom, kao kontrolnim materijalom, bila prisutna reakcija odbacivanja. Na osnovu dobijenih rezultata može se zaključiti da se ispitivani biokeramički materijali mogu koristiti za popunjavanje koštanih defekta u ortopedskoj hirurgiji i stomatologiji.",

The epithelium of the periodontal pocket--an electron microscopy analysis

AB  - The aim of this study was to examine intra- and intercellular changes that characterize the periodontal pocket epithelian. Biopsy specimens of human gingiva were prepared for an electron microscope study by a routine histological procedure. Results of the histological preparations analyses revealed changes significant for pathogenesis of gingivitis and periodontal disease because they cause destruction of epithelial barrier. Such a damaged periodontal pocket epithelium enables further invasion of harmful noxae and spreading of destructive processes into deeper parts of the periodontium.
